摘  要:目的探讨顺铂联合脱氧氮杂胞苷对三阴性乳腺癌细胞增殖及凋亡的影响。方法以人三阴性乳腺癌细胞MDA-MB-231为研究对象,分为A、B、C组。A组:MDA-MB-231细胞用5.0μmol·L-1脱氧氮杂胞苷进行处理48 h;B组:MDA-MB-231细胞用10μmol·L-1顺铂进行处理48 h;C组:MDA-MB-231细胞用5.0μmol·L-1脱氧氮杂胞苷联合10μmol·L-1顺铂进行处理48 h。用噻唑蓝法测定3组细胞处理后的增殖抑制率,用流式细胞仪测定3组细胞处理后的凋亡比例。结果 3组细胞随培养时间的增加,增殖抑制率明显增高(P<0.05),C组增殖抑制率显著高于A,B组(P<0.05)。3组细胞随培养时间的增加,凋亡比例明显上升(P<0.05),C组细胞凋亡率显著高于A,B组(P<0.05)。结论顺铂联合脱氧氮杂胞苷可明显抑制三阴性乳腺癌细胞的增殖,促进其凋亡。Objective To investigate the effect of cisplatin combined with deoxy-azacytidine(DAC) on proliferation and apoptosis of triple-negative breast cancer cell.Methods Triple-negative breast cancer cells MDA-MB-231 for the study were divided into A,B,C group.Group A:MDA-MB-231 cells were treated with 5.0 μmol·L-1DAC processing 48 h.Group B:MDA-MB-231 cells were treated with 10μmol·L-1cisplatin treatment 48 h.Group C:MDA-MB-231 cells48 h treatment with 5.0 μmol·L-1DAC combined with 10 μmol·L-1cisplatin.The cell proliferation rate and apoptosis rate were measured by MTT and flow cytometry flow cytometry assay.Results The cell proliferation was significant inhibit in three groups(P 〈 0.05) with the group C higher than group A and B(P 〈 0.05).And the apoptosis rate in group C was significant higher than group A and B(P 〈 0.05).Conclusion Cisplatin combined with deoxy-azacytidine can inhibit the proliferation and promote the apoptosis of MDA-MB-231 human breast cancer cell line.
