摘 要:目的:对胡黄连苷-Ⅱ原料药进行结构鉴定,建立HPLC含量测定方法。方法:根据理化常数和光谱数据进行结构鉴定;采用反相高效液相色谱法,对胡黄连苷-Ⅱ原料药进行含量测定。结果:胡黄连苷-Ⅱ原料药结构鉴定表明其成分为6-香草酰基梓醇,即胡黄连苷-Ⅱ;HPLC测定胡黄连苷-Ⅱ在0.315~5.05μg进样量与峰面积线性关系良好,平均加样回收率为98.09%,RSD 1.6%,3批胡黄连苷-Ⅱ原料药质量分数分别为101.53%,101.50%,102.25%。结论:胡黄连苷-Ⅱ原料药含量较高,所建立含量测定方法简便可行,重复性好,可用于胡黄连苷-Ⅱ原料药的质量控制。Objective: To identify the structure of picroside-Ⅱ and establish an HPLC method for determining the content of picroside-Ⅱ . Method: The structure was elucidated by its physiochemical characteristics and spectral analysis. The content of picroside-Ⅱ was determined by HPLC. Result: The structure was elucidated as 6-vanilloyl catalpol ( picroside- Ⅱ ). The linear range of picroside- Ⅱ was within 0. 315-5.05 txg. The average recovery was 98.09% , with RSD 1.6%. The three batches of active pharmaceutical ingredient picroside-Ⅱ were 101.53%, 101.50%, and 102.25%, respectively. Conclusion: The method is simple and specific, and can be used for control of picroside- Ⅱ.
