摘 要:目的探讨融合基因(TMPRSS2-ERG)阳性前列腺癌中TOMM34基因的表达及其意义。方法采用实时荧光定量PCR法和Western Blot法检测TOMM34在前列腺癌细胞株LNCAP(ERG阴性)、PC-3(ERG阴性)及VCAP(ERG阳性)中的表达情况。同时采用免疫组化方法检测ERG及TOMM34在前列腺组织中的表达。结果 TOMM34在前列腺癌细胞株VCAP中的m RNA的表达水平要明显低于其在PC-3[(0.91±0.10)VS(2.30±0.59),P<0.05]及LNCAP[(0.91±0.10)VS(4.00±0.22),P<0.05]的表达水平,且其在VCAP中的蛋白表达水平也要明显低于PC-3及LNCAP中的表达水平(P<0.05)。免疫组化结果提示在ERG阳性的前列腺癌组织中,TOMM34的阳性率为25%(4/12),而在ERG阴性前列腺癌组织中其阳性率为81.8%(18/22),(χ2=12.268,P<0.001)。ERG在前列腺癌组织中蛋白的表达水平与患者Glesason评分呈负相关性(P<0.05),TOMM34在前列腺癌组织中蛋白的表达水平与患者Glesason评分呈正相关性(P<0.05)。结论 TMPRSS2-ERG基因融合阳性前列腺癌组织中TOMM34表达明显低于TMPRSS2-ERG基因融合阴性前列腺癌。Objective To investigate the differences and significance of TOMM34 gene expression between TMPRSS2-ERG fusion gene-positive and TMPRSS2-ERG fusion gene-negative prostate cancer. The expression of TOMM34 in three prostate cancer cell lines (ERG positive: VCAP; ERG negative: LNCAP, PC-3) was determined by RT-PCR and Western Blot. Meanwhile, the expression of TOMM34 and ERG protein in 38 cases of prostate cancer was detected by immunohistochemistry. The expression of TOMM34 gene mRNA and protein were significantly different between prostate cancer cell types. Its expression decreased in ERG-positive prostate cancer cells (VCAP), while it was increased in ERG-negative prostate cancer cell (LNCAP, PC-3)(P〈0.05). The results of immunohistochemistry showed that the TOMM34 expression level in ERG-positive prostate cancer was significantly lower than that in ERG-negetive prostate cancer. In addition, we found that the level of ERG in prostate cancer were inverse correlated with Gleason Grade (P〈0.05), and TOMM34 was positive correlated with Gleason Grade (P〈0.05). Conclusion There is a significant inverse correlation between the expression of ERG and TOMM34 in both prostate cell lines and cancer tissues.
