摘  要:目的探讨肝星型细胞(Hepatic stellate cell,HSC)的表达与上皮-间质转化(epithelial-mesenchymal transition,EMT)的关系,并结合临床资料分析HSCs对原发性肝癌(primary hepatocelluar carcinoma,HCC)生物学行为的影响。方法通过免疫组织化学方法检测30例肝细胞癌癌组织及23例癌旁组织中的活化HSCs的特异性标记蛋白α-SMA及Twist,同时结合临床资料进行分析。结果肝细胞癌癌组织及癌旁组织中,α-SMA阳性表达率分别为60.0%和30.4%,组间差异有统计学意义(P<0.05);Twist阳性表达率分别为66.7%和39.1%,组间差异有统计学意义(P<0.05)。肝癌组中α-SMA和twist阳性表达组和阴性表达组之间的肿瘤分化程度和分期差异有统计学意义(P<0.05)。30例肝癌病理标本中,α-SMA与twist蛋白表达呈正相关(r=0.433,P<0.05)。结论 HSCs在HCC中高表达,对评估肝癌EMT及预后具有重要意义。Objective To evaluate the correlation between the biomarkers expression of hepatic stellate cells(HSCs)and epithelial-mesenchymal transition(EMT) of hepatocellular carcinoma(HCC)and analyze the relationship between HSCs and clinical/pathological factors and the prognosis. Methods Expression of α-SMA and Twist in 30 HCCs and 23 pericancerous tissues was detected by SP immunhistochemical method. Correlation between α-SMA and Twist of HCC as well as clinical/pathological parameters and prognosis was analyzed. Results In HCCs and pericancerous tissues, the positive expression rates of α-SMA were 60.0% and 30.4%(P〈0.05), and those of Twist were 66.7% and 39.1%(P〈0.05), respectively. According to α-SMA expression in HCC tissue,the clinical/pathological indexes of the tumor differentiation and stage were statistically correlated with α-SMA.The expression of α-SMA and Twist was positively correlated in HCC(r=0.4339, P〈0.05). ConclusionExpression of SMA and Twist in HSC may be pivotal in evaluation of epithelial-mesenchymal transformation and prognosis in HCC.
