摘 要:目的通过观察可溶性IL-5受体α(sIL-5Rα)对支气管哮喘(简称哮喘)小鼠气道炎症的影响,为哮喘疾病的治疗提供新的方法。方法24只BALB/c小鼠随机分成正常组、哮喘组及sIL-5Rα治疗组,哮喘组和sIL-5Rα治疗组以鸡卵白蛋白(OVA)致敏和激发,建立哮喘模型,对照组用生理盐水代替0VA。sIL-5Rα治疗组每次激发前30min腹腔注射sIL-5Rα100μg,正常对照组及哮喘组则用生理盐水替代。比较各组BALF细胞计数,IL-5、IL-4及嗜酸粒细胞趋化因子(Eotaxin)水平,肺组织学检查。结果与正常对照组相比,哮喘组BALF中嗜酸粒细胞百分比增加(P〈0.05),BALF中IL-4、IL-5、Eotaxin水平增加(P〈0.05),病理示肺组织损害明显。与哮喘组相比,sIL-5Rα治疗组BALF中嗜酸粒细胞百分比降低(P〈0.05),BALF中IL-5、Eotaxin水平增加(P值均〈O.05),病理示肺组织损害明显减轻。结论sIL-5Rα可明显减轻哮喘小鼠气道炎症。Objective To investigate the therapeutic potency of soluble interleukin-5 receptors (sIL-5Rα) in bronchial asthma (asthma) and determine the influences of sIL-5Rα on airway inflammatory response insensitized mice. Methods Twenty-four mice were devided randomly into three groups: control group,asthmatic group and sIL-5Rα group. Mice in the latter two groups were sensitized and challenged with ovalbumin (OVA) to establish asthmatic models, while mice in control group were treated with saline as the substitution of OVA. Mice in treatment group were injected intraperitoneally with the sIL-5Rα (100 μg) 30 min before challenged, while mice in control group and asthmatic group received saline. Histological examinations were undertaken. Cell accounts and cytokines (IL-5, IL-4, Eotaxin) of the bronchoalveolar lavage fluid (BALF) were evaluated. Results Compared with control group,total cell and eosinophil percentage of BALF, IL-5 and IL-4 in BALF and were all increased considerably after OVA challenge. The infection of lung issue was significant. Compared with asthmatic group, the above changes was decreased significantly in sIL-5Rα group. Conclusions sIL-5Rα could significantly alleviate airway inflammation of the asthma mouse.
