丰富环境对缺氧缺血新生大鼠神经行为能力及脑髓鞘形成的影响

摘  要:目的探讨不同干预起始时间和强度的丰富环境对缺氧缺血性脑损伤(HIBD)新生大鼠神经行为能力及髓鞘形成的影响。方法建立HIBD新生大鼠的丰富环境干预模型,分别在缺氧缺血(HI)术后第7、14、21天(即早、中、晚期)开始丰富环境干预,持续14d,每天干预6h或24h。以前肢悬吊试验和水迷宫试验检测大鼠神经学预后,以免疫组织化学法检测脑白质髓鞘碱性蛋白(MBP),用图像分析仪测定阳性细胞在视野中的百分比。结果神经行为学测试结果显示,早中期假手术组、早中期HI组、早期6h干预组、早期24h干预组、中期6h干预组、早期24h干预组、晚期假手术组、晚期HI组、晚期24h干预组悬吊试验成绩分别为(4.05±0.88)分、(2.35±1.02)分、(3.67±1.12)分、(3.50±1.41)分、(3.50±0.93)分、(3.56±1.13)分、(4.00±0.89)分、(2.17±1.17)分和(3.50±0.92)分,早中晚期各干预组悬吊试验成绩均较同期HI组显著提高,早中期6h干预组和24h干预组之间成绩差异无统计学意义。而相应组别的水迷宫试验成绩分别为(40.68±23.77)s、(56.66±10.96)s、(46.49±19.27)s、(51.72±20.46)s、(38.20±18.36)s、(47.96±20.65)s、(38.63±20.44)s、(59.66±13.81)s和(45.93±22.45)s,早中期6h干预组以及晚期24h干预组水迷宫试验成绩显著高于同期HI组,而早中期6h干预组和24h干预组之间成绩差异无统计学意义。免疫组织化学检测结果显示,早中期HI组和晚期HI组白质MBP相对含量为6.32±1.63和6.74±2.19,显著低于同期假手术组(9.09±1.69,9.37±2.46)。早期6h、24h干预组,中期6h、24h干预组,晚期24h干预组的白质MBP相对含量分别为7.84±2.51、8.05±1.86、8.89±2.29、8.48±2.67、7.98±2.09,显著高于同期HI组。结论�Objective To explore the impactof enriched environmen(EE), which hadiffereninitiation time pointand intensity, on the neural and ethological prognosiand contentof myelin basiprotein(MBP) of neo- natal ratwith hypoxi- ischemibrain damage (HIBD). MethodHIBD ramodelwere established. Ratwere divided into the early, the intermediate and the late intervention groups, which experienced EE from 7, 14 and 21 dayafteHIBD fo14 days. The early and intermediate intervention groupwere then divided into 6 - h and 24 - h groups, which experienced EE intervention fo6 houro24 hourrespectively each day. Trapeze testand watemaze testwere carried outo detecthe neural and ethological prognosis. Immunohistochemical method waused to detecMBP of the brain white matter, and the percentageof positive cellwith MBP were detected by an image analyzer. The con- tentof MBP were measured. ResultThe trapeze tesscoreof the early and intermediate sham operation group, HI group, early 6 - h and 24 - h EE groupand the intermediate 6 - h and 24 - h EE groups, the late sham operation group, the late HI and late EE intervention group were (4.05 ±0.88) scores, (2.35 ± 1.02) scores, (3.67 ± 1.12) scores, (3.50 ± 1.41) scores, (3.50 ± 0.93) scores, (3.56 ± 1. 13 ) scores, (4.00 ± 0.89) scores, (2. 17 ± 1.17) scores, (3.50 ± 0.92) scores, respectively. The trapeze tesscoreof early, intermediate and late EE groupwere highethan those of the HI groupin the same period. There wano significandifference between the early, the intermediate 6 - h EE groupand 24 - h EE groups.
