摘 要:分离马尾杉内生真菌,并筛选其具抑菌活性菌株。用组织块分离法分离纯化菌株,通过形态学观察对其进行初步鉴定,以大肠杆菌、枯草芽孢杆菌和金黄色葡萄球菌为指示菌,采用对峙法进行抑菌活性筛选。实验分离获得马尾杉内生真菌28株,其中叶4株,叶鞘12株,茎9株,根3株。革兰氏染色结果表明,28株菌中,17株菌的菌丝染色呈紫色(60.71%),11株菌染色呈红色(39.29%),各菌的菌丝形态和孢子形态各异。抑菌实验结果表明所得28株马尾杉内生真菌,有11株至少能对1种指示菌有抑菌作用,占总分离菌株的39.29%,其中3株对金黄色葡萄球菌有较强的抑菌活性。马尾杉中存在大量的内生真菌,有些内生菌能产生抑菌活性代谢产物,可以进一步对这些代谢产物开展研究,为微生物药物资源的开发提供前期的物质基础。The purpose is to isolate the endophytic fungi from Phlegmariurusphlegmaria Holub and to screen the antibacterial activity of these strains. Endophytic fungi were isolated with tissue inoculation culture and identified by conventional morphological characteristic method. Escherichia coli,Bacillus subtilis and Staphylococcus aureus were used as indicators to do the microbial inhibition experiment. A total of 28 strains of endophytic fungi were isolated,including 4 strains from leaf,12 strains from sheath,9 strains from stem and 3 strains from root. Gram staining results showed that 17 strains were purple( 60. 71% of the total strains isolated) and,11 strains were red( 39. 29% of the total strains isolated). Mycelial and spore of endophytic fungi are of different forms. The antibacterial test showed that,among 28 strains of endophytic fungi isolated from Phlegmariurusphlegmaria Holub,11 strains( 39. 29%of the total strains isolated) showed the antibacterial activity against at least one of indicator bacteria. And 3 stains of them showed the stable and strong antibacterial activity against Staphylococcus aureus. The Phlegmariurusphlegmaria Holub contains abundant endophytic fungi,some of which can produce antibacterial active metabolites. Further research can be focused on these metabolites,which may provide the material basis for microbial medicine resource development.
