摘  要:目的建立电感耦合等离子体发射光谱测定母乳中钙磷含量的方法,并对深圳市当地母乳钙、磷两种元素的含量进行测定。方法于2011—2013年收集深圳市产后1~2个月产妇的母乳样品共221例,样品冻干保存,经硝酸高压消解后采用电感耦合等离子体发射光谱法(ICP一0Es)测定母乳中钙和磷的含量。结果钙检出限0.09roe,/kg,磷检出限0.03me./kg;各元素线性良好,相关系数R〉0.9999;钙测定精密度1.3%~2.5%,磷测定精密度1.6%~2.1%;国家标准参考物质的分析结果在证书值范围内;深圳市母乳中钙含量187.6~449.6mg/kg,平均值为294.5me,/kg,磷含量84.9--230.7me./kg,平均值为157.7me./kg,钙磷比1.1~3.7,平均值为1.9。结论电感耦合等离子体发射光谱法灵敏、准确可靠,可用于母乳中钙磷元素的同时检测。深圳市母乳中钙元素的平均含量在WHO提供的参考值范围内,磷元素的平均含量略高于WHO提供的参考值范围。Objective To develop a method for determination of calcium (Ca) and phosphorus (P)in human milk, and to in- vestigate the contents of Ca and P in human milk in Shenzhen City. Methods A total of 221 human milk ( 1 - 2 months postpartum) samples were collected and preserved after lyophilization in Shenzhen City during the period of 2011 - 2013. After high- pressure digestion by HNO3 in a closed - vessel, the dements of Ca and P were simultaneously determined by inductively coupled plasma optical emission spectrometer (ICP - OES). Results The detection limits of Ca and P were 0.09 mg/kg and 0.03 mg/kg respectively. The correlation coefficients were both ~0.9999, with a good linearity. The relative standard deviation of Ca and P ranged from 1.3 % to 2.5 % and from 1.6 % to 2.1% respectively. The detection results of international reference standard materials for Ca and P both fell in the certificate value ranges. The level of Ca in human milk in Shenzhen City ranged from 187.6 mg/kg to 449.6 mg/kg, and the average concentration was 294.5 mg/kg. The level of P ranged from 84.9 m_g/kg to 230.7 mg/kg, and the average concentration was 157.7 mg/kg. The ratio of Ca and P ranged from 1.1 to 3.7, with the average value of 1.9. Conclusions The inductively coupled plasma optical emission spectrometer (ICP- OES) has a high accuracy and good reproducibility, which can be used to simultaneously determine the elements of Ca and P in human milk. The average content of Ca in human milk in Shenzhen is within the current reference ranges provided by the World Health Organization (WHO), while the average content of P slightly exceeds the current reference ranges provided by the WHO.
