摘  要:目的:观察不同提取工艺对九香虫中脂肪油和氨基酸成分的影响,为九香虫的综合利用提供实验依据。方法:分别采用石油醚溶剂回流提取法和超临界流体CO2萃取法提取九香虫脂肪油;经甲酯化处理后,采用GC-MS对脂肪油成分进行分析。并对两种工艺提取后的残渣用盐酸水解后,利用氨基酸自动分析仪对其所含氨基酸进行分析。结果:溶剂回流提取脂肪油的得率为43.39%,从中鉴定出14个成分,已鉴定成分含量占总量的58.49%,不饱和脂肪酸的含量为39.03%。超临界流体CO2提取脂肪油的得率是42.23%,并从中鉴定出17种成分,已鉴定成分含量占总量的95.26%,不饱和脂肪酸的含量为74.58%。超临界流体提取后残渣中的总氨基酸含量为48.7%,粗蛋白含量为56.1%;而溶剂回流提取处理后残渣中的总氨基酸含量是55.5%,粗蛋白是61.1%。结论:不同提取工艺所得九香虫中脂肪油和氨基酸不仅存在量的差异,其内在成分也明显不同,因此还应结合药效学考察确定九香虫的提取工艺。Objective : To investigate the effect of fatty oil and amino acid by different extraction technologies, and to provide experimental evidence for development of Aspongopus. Method : With petroleum ether solvents refluxing extraction and supercritical fluid CO2 extraction method to extract the fatty oil in Aspongopus, it was modified to methylester and then it was determined by GS - MS. And two kinds extraction residue were processed with hydrochloric acid and amino acids containments were analyzed with automatic amino acid analyzer. Results:The fatty oil extracted by petroleum ether reflux was 43.39%. 14 components were identified, accounting for 58.49%. The content of unsaturated fatty acid was 39. 03%. The fatty oil extracted by SFC was 42.23%, 17 components were accounting for 95.26%. The content of unsaturated fatty acid was 74.58%. Conclusion:There were not only quantity difference between Aspongopus fats oils and amino acids got from two kinds extraction processes, but also differences in its inherent components, and therefore pharmacodynamic study should be combined to determine the extraction process of Aspongopus.
