摘  要:目的探讨血管上皮生长因子(VEGF)、色素上皮衍生因子(PEDF)在不同病程糖尿病大鼠视网膜中的表达情况。方法将雄性SD大鼠120只按照随机数字表格法划分为正常对照组与糖尿病组,每组60只。将糖尿病大鼠按照一次性腹腔注射链脲佐菌素方法制作成糖尿病模型。成模后1,3,5个月分别应用透射电镜切片、视网膜EB含量测定法观测大鼠视网膜的病理变化情况,同时应用Western-blot技术以及免疫组化染色测定糖尿病大鼠视网膜中VEGF、PEDF表达情况。结果 EB染色显示正常对照组大鼠的视网膜血管形态较好;1个月时糖尿病大鼠视网膜血管较规则;3个月时糖尿病大鼠视网膜血管迂曲,有部分改变;成膜后5个月时糖尿病大鼠视网膜血管迂曲已经十分明显,且走行不规则,血管粗细不均。同时成模后1,3,5个月糖尿病组大鼠EB含量均高于正常对照组(P均<0.05),且随病程进展,其含量也增加。免疫组化显示糖尿病大鼠VEGF的阳性染色会随病程进展而有所增强,光密度值增大;PEDF阳性染色则随病情进展而减弱,同时光密度值减小;与正常对照组比较差异均有统计学意义(P均<0.05)。Western-blot法检测显示随着病情进展,糖尿病组各阶段大鼠的视网膜VEGF蛋白含量随之增加,而PEDF蛋白则减少,与正常对照组比较差异均有统计学意义(P均<0.05)。结论最早可观察到的视网膜病变是视网膜细胞超微结构病理变化,会随病情进展加重;视网膜血管形态在病情中后期会发生变化,且随病情加重而加重;在初期,视网膜的VEGF与PEDF表达已经失衡,且随病情进展,VEGF表达增加,PEDF表达减少,二者失衡贯穿于整个病情的发展过程中。Objective It is to observe the expression of vascular endothelial growth factor (VEGF) and pigment epitheli- um-derived factor (PEDF) in the retina of different course of diabetic rats. Methods 120 cases of male outbred rats (SD rats) were selected to divide into the normal control group and diabetes group according to the random number table method as 60 cases in each. The diabetic rat model was made by intraperitoneal injection of streptozotocin method. After 1, 3, and 5 month, the retinal pathological changes were observed by transmission electron microscope and retinal EB content determina- tion method respectively, and the expression of VEGF and PEDF in the retina of diabetic rats were observed by Western-blot technology and immunohistochemical methods. Results The EB staining showed of the retinal vessels morphology in rats of nor- mal control group was good, the retinal vessels was regular in the first month, and they was tortuose with some changes in the third month; After modeling success in the fifth month, the tortuosity had been very clear, and go for irregular with uneven thickness of vascular. At the same time, the EB content in rats of diabetic group in the 1 st, 2nd, and 3rd month after model- ing successful were significantly higher than that of normal control group ( all P 〈 0.05) , and content was increased with the progress of the disease. Immunohistochemistry showed, the positive staining of VEGF in diabetic rats was increased with dis- ease progression, the value of optical density was increased ; the positive staining of PEDF was increased with the progress of the disease and weakened, and the value of optical density was decreased. There were significant differences between the con- trol group and diabetic group ( all P 〈 0.05 ). Western-blot assay showed that the retinal VEGF protein content in each stage of diabetic group were increased with the progress of the disease, and the PEDF protein were decreased, there was significant difference between control group and
