摘 要:目的:观察不同葡萄糖浓度下大鼠心肌细胞H9C2的糖代谢变化,初步探讨糖毒性对心肌细胞的损伤作用。方法:以体外培养的大鼠心肌细胞系H9C2为模型,观察不同糖浓度下,心肌细胞的形态学变化,并通过MTT实验检测存活率,流式细胞仪检测H9C2细胞凋亡率。结果:培养24 h后,H9C2细胞形态开始发生变化;培养48 h后,H9C2细胞存活率明显下降、凋亡率上升,且与葡萄糖浓度相关。45 mmol/L葡萄糖刺激心肌细胞48 h后,心肌细胞存活率降至(45.09±0.84)%;持续刺激72 h后,细胞死亡率进一步增高,心肌细胞存活率降至(37.93±0.66)%。15 mmol/L和45 mmol/L葡萄糖处理心肌细胞48 h以上,细胞存活率与对照组相比均显著下降。结论:葡萄糖浓度升高和作用时间延长可促进心肌细胞发生凋亡,高糖对心肌细胞有直接损伤作用。Objective:To study the glucose metabolism state in H9C2 myocardial cell lines under different concentrations of glucose. Methods:H9C2 myocardial cells were cultured in vitro and used as a model system.Morphological changes of myocardial cells were observed under different concentrations of glucose,and the survival rate was measured by MTT assay.Flow cytometry was used to investigate the apoptosis rate. Results:After 24-hour culture under different concentrations of glucose,the myocardial cells began to change in morphology.Forty-eight hours after culture,the survival rate decreased and the apoptosis rate increased,which indicated a dose-dependent property.After stimulation with 45mmol/L glucose for 48 h,the cells survival rate decreased to(45.09±0.84)%,and reached(37.93±0.66)%after stimulation for 72 h.Glucose-stimulation at concentrations of 15mmol/L and 45mmol/L for over48 hboth led to decreased cells survival rate compared with the controls. Conclusion:Apoptosis is induced in myocardial cells with the increasing concentrations and prolonged stimulation of glucose.High glucose causes direct damage to myocardial cells.
