摘 要:以芦丁为对照品,选择510 nm为测定波长,580 nm为参比波长,采用双波长分光光度法测定洋葱中黄酮含量。试验结果表明,芦丁在0.008~0.040 mg/mL质量浓度范围内,与吸光度差值ΔA呈良好的线性关系,回归方程为ΔA=5.95C+0.007 3,相关系数r=0.998 9,检出限为0.075 mg/L,测定方法的平均回收率为99.79%,RSD=1.30%。测得洋葱中黄酮含量为1.58 mg/g。试验建立的方法操作简便,稳定可靠,为洋葱的开发和检测提供了依据。Used Rutin as a standard material, a double-wavelength spectrophotometry was proposed for the onion flavonoid determination, the chosen detection wavelength were 510 nm and 580 nm as the reference. The result showed a good linearity with absorbance (AA) at the Rutin concentration range of 0.008-0.040 mg/mL, the regression equation was AA=5.95C+0.007 3, with the correlation coefficient of 0.998 9, the detection limit was 0.075 mg/L, and the average recovery was 99.79% (RSD=1.30%). The onion flavonoid content was 1.58 mg/g detected by the method. It improved the accurate and reliable offlavonoid detection, and provided the basis for the development and determination of onion.
