周期性应力条件下整合素过表达对大鼠软骨细胞的影响

摘  要:目的探讨周期性应力条件下整合素过表达对大鼠软骨细胞的影响。方法体外分离培养大鼠软骨细胞,随机分为上调组(A组,整合素过表达质粒的慢病毒转染)和对照组(B组,空白质粒的慢病毒转染)。将细胞置于周期性应力场中培养,采用Western blot法检测细胞外调节蛋白激酶1/2(ERK1/2)蛋白表达和磷酸化水平,实时定量荧光PCR检测蛋白聚糖和Ⅱ型胶原基因表达水平,细胞计数试剂盒(CCK-8)检测软骨细胞增殖情况。结果 A组整合素表达高于B组(0.824±0.029vs.0.419±0.014)(P<0.05)。与B组相比,在周期性应力作用下,A组ERK1/2磷酸化水平、蛋白聚糖和Ⅱ型胶原基因表达、细胞增殖均增加(P<0.05)。结论整合素过表达可以促进周期性应力作用下软骨细胞的增殖和基质合成。Objective To investigate the effect of integrin overexpression under periodic mechanical stress on chondroctyes in rats .Methods Chondrocytes were cultured in vitro and then randomly divided into two groups of A (transfected with recombinant plasmid containing integrin overexpression) and B (transfected with empty plasmid ) .After chondrocytes were cultured under periodic mechanical stress or static condition ,the protein expression and phosphorylation level of ERK1/2 were detected by Western blot .The gene expressions of aggrecan and typeⅡ collagen were determined by qRT‐PCR ,and cell proliferation was measured by CCK‐8 assay .Results Integrin expression in group A was higher than that in group B(0.824 ± 0.029 vs .0.419 ± 0.014)(P〈0 .05) . Compared with group B ,the phosphorylation level of ERK1/2 ,gene expressions of aggrecan and typeⅡ collagen and cell proliferation were all increased under periodic mechanical stress in group A (P〈0 .05) .Conclusion Integrin overexpression can promote the proliferation and matrix synthesis of chondrocytes under periodic mechanical stress .
