疣状胃炎病损组织与同遗传背景胃癌组织的差异蛋白组学研究

摘　　要：[目的]通过差异蛋白组学方法分析疣状胃炎与同遗传背景胃癌组织的差异蛋白表达,旨在筛选该型胃炎向胃癌转变过程中关键的蛋白分子,探讨其癌变的分子机制。[方法]外科手术获取内镜及病理检查提示同时存在胃癌与疣状胃炎病变患者的病变组织,采用比较蛋白组学通用的双向电泳及质谱分析技术,筛选并鉴定胃癌组织及癌周疣状胃炎组织中差异表达的蛋白质。[结果]胃癌组织与癌旁疣状胃炎组织比较,共筛选出显著差异表达蛋白点34个,其中表达上调蛋白质点20个,表达下调蛋白点14个;共有27个蛋白点被鉴定,上调蛋白17个,下调蛋白10个。[结论]在疣状胃炎基础上发生的胃癌与其疣状胃炎病损组织的蛋白质表达存在显著差异,这些差异表达蛋白分子涉及细胞增殖、凋亡、运动、物质能量代谢等诸多功能领域,其代表的分子信号转导途径可能揭示疣状胃炎癌变的分子机制。[Objective]To investigate the molecular mechanism underlying the transform form varioliform gastritis（VG）to gastric cancer by a differential proteomic study to screen the key proteins in the transform. [Methods]A combination of two-dimensional gel electrophoresis and mass spectrometry was conducted to analyze the differentially expressed proteins between gastric cancer lesions and varioliform gastritis. [Results]Thirty-four differentially expressed proteins were found and 27 differentially expressed proteins were identified in gastric cancer,compared with matched VG mucosa,17 proteins were upregulated and 10 downregulated in gastric cancer when compared with the same proteins in individual-matched VG mucosa. [Conclusion]Some difference of protein expression between gastric cancer and VG mucosa were demonstrated and the function of these proteins could be related to cell proliferation,poptosis and as so on. The differentially expressed proteins might play some important roles in cancerization from VG as functional molecules.
