摘  要:趋化因子受体CCR5(CC chemokine receptor 5)是G蛋白偶联受体超家族的一员,主要表达在机体多种免疫细胞表面。CCR5依赖与其特异性配体的结合而发挥生物学作用,在细胞的生长、活化、分化、粘附及定向迁移等方面起着重要作用。CCR5是人免疫缺陷病毒1型(human immunodeficiency virus type-1,HIV-1)入侵人体的重要辅助受体之一,参与病毒与T细胞结合的过程。靶向CCR5的治疗不仅具有抗病毒作用,还具有免疫调节作用。研究发现,CCR5不仅在AIDS、自身免疫病、动脉粥样硬化等疾病的致病机制中起作用,在异基因造血干细胞移植(allogeneic hematopoietic stem cell transplantation,allo-HSCT)后急性移植物抗宿主病(acute graft versus host disease,a GVHD)发生发展过程中,同样扮演着重要的角色。大量动物实验证实,CCR5及其配体介导了效应T细胞向GVHD靶器官的迁移与募集。因此,本文就CCR5的结构和功能、内化途径、信号转导通路及其与allo-HSCT后a GVHD的关系作一综述。Chemokine receptor-5 ( CCR5 ) belongs to a G-protein coupled receptors superfamily. It is mainly expressed on a wide variety of immune ceils. CCR5 can bind with its specific ligands, which plays very important roles in inflammatory cell growth, differentiation, activation, adhesion and migration. CCR5 was identified as a co-receptor for human immunodeficiency virus type-1 (HIV-1) to infect CD4+ T cells. In addition, CCR5 not only participates in the pathogenic mechanisms of many inflammation disease such as AIDS, auto-immune disease, and atherosclerosis, but also plays important roles in the development of acute graft-versus-host disease after allogeneic hematopoietic stem cell transplantation. Recent studies using murine models have demonstrated the critical role of CCR5 and its ligands which direct T-cell infiltration and recruitment into target tissues during acute GVHD. CCR5 has become the focus of intense interest and discussion, and this review will attempt to describe what is understood about the structure and function, internalization, signal transduction of CCR5, in order to investigate the relationship between CCR5 and acute GVHD.
