摘 要:目的探讨再生室内联合应用牙髓干细胞(dental pulp stem cells,DPSCs)和转化生长因子-β3(transforming growth factor-β3,TGF-β3)修复兔面神经横断性损伤的可行性,比较各时间段不同药物干预后面神经的恢复情况。方法健康成年新西兰大白兔36只,随机分为DPSCs+TGF-β3组(实验组)、TGF-β3组和PBS组,每组各12只,3组均于兔左面颊部分离面神经上颊支并切断,利用硅胶管作为再生室桥接离断的神经上颊支,建立面神经上颊支横断伤模型。实验组在再生室内加入100ng/μL TGF-β3液和1×108/LDPSCs悬液0.1mL,TGF-β3组加入等量100ng/μL TGF-β3液,PBS组加入等量PBS液,术后4、12周比较3组兔面神经修复再生情况。结果4、12周时实验组神经纤维数[(1 034.58±94.28)、(1 425.92±188.98)根]和纤维直径[(2.05±0.14)、(7.18±0.40)μm]高于TGF-β3组[(935.00±44.60)、(1 053.42±135.26)根,(1.56±0.12)、(6.21±0.47)μm]和PBS组[(555.50±80.20)、(694.36±24.11)根,(1.26±0.08)、(3.64±0.56)μm](P<0.05);12周时3组再生神经纤维总数和纤维直径均高于4周时(P<0.05)。结论 TGF-β3与DPSCs联合应用可有效促进面神经再生,二者联合应用较单用TGF-β3修复效果好;DPSCs可作为种子细胞应用于面神经组织工程。Objective To investigate the feasibility of combined application of transforming growth factor-β3 (TGF-β2) and dental pulp stem cells (DPSCs) in repairing rabbit facial nerve transverse trauma, and to compare the recovery of facial nerve after intervention of different drugs in different periods of time. Methods Thirty-six healthy adult New Zealand rabbits were randomly divided into DPSCs+TGF-β3 group, TGF-β3 group and PBS group, with 12 rabbits in each group. All rabbits were separated and cut off the buccal branch of facial nerve at the left cheek. Silicone tube was used as regeneration chamber to make a bridging joint on isolated buccal branch of nerve to establish models of traumatic transection on buecal branch. DPSCs+TGF-β3 group was added 100 ng/μL TGF-β3 fluid and 0. 1 mL 1 × 10^8/LDPSCs suspension into regeneration chamber, TGF-β3 group was added the same amount of 100 ng/μL TGF-β3 fluid, and PBS group was added the same amount of PBS fluid. The recovery of facial nerve regeneration was compared among three groups in 4 and 12 weeks after operation. Results The number of nerve fibers (1 034.58±94.28, 1 425.92±188.98) were larger and fiber diameter ((2.05±0.14), (7.18±0.40)μm) were longer in experimental group in 4 and 12 weeks than those inTGF-β3 group (935.00±44. 60, 1 053.42±135.26; (1.56±0.12), (6.21±0.47) μm) and PBSgroup (555. 50±80.20, 694.36±24.11% (1.26±0.08), (3.64±0. 56) μm) (P〈0.05). The total number of regeneration nerve fibers and fiber diameter were larger and longer in all three groups in 12 weeks than those in 4 weeks (P〈0.05). Conclusion The combined application of TGF-β3 and DPSCs can effectively promote the facial nerve regeneration, with a better effect than single application of TGF-β3. DPSCs could be applied in facial nerve tissue engineering as seed cells.
