摘  要:目的探索低频电刺激脚桥核(PPN)对偏侧6-羟基多巴胺(6-OHDA)毁损帕金森病(PD)大鼠丘脑腹外侧核(VL)神经递质水平的影响。方法 27只雄性大鼠随机分为假损毁组A(n=8)、损毁组B(n=9)和损毁+电极组C(n=10)。动物行离体定向手术后,进行微透析取样,对样品用高效液相和液相质谱(LC-MS)连用的方法检测神经递质浓度。结果 6-OHDA损毁后,大鼠VL内谷氨酸(Glu)和γ-氨基丁酸(γ-GABA)水平显著升高,乙酰胆碱(Ach)轻微下降;低频刺激PPN期间,Glu浓度显著降低,ACh轻微升高,γ-GABA没有变化。结论低频电刺激PPN可再平衡PD大鼠VL内Glu及Ach浓度,该作用可能与其改善PD大鼠姿势不稳和步态困难的机制有关。Objective The effects of low frequency stimulation of pedunculopontine nucleus ( PPN ) on neurotransmitter of ventrolateral thalamic nucleu (VL) in unilateral 6-hydroxy dopamine (6-OHDA)-lesion rats are investigated.Methods Twenty-seven male SD rats were randomly assigned to sham-lesion group A (n=8), lesion group B (n=9), and lesion+electrodes group C (n=10).After stereotactic surgery, these samples were collected by microdialysis.The neurotransmitter in every sample was detected by high performance liquid chromatography and mass spectrometry (LC-MS).Results The 6-OHDA lesion resulted in a significant increase of the level of the glutamic acid (Glu) andγ-aminobutyric acid (γ-GABA) in VL, but a little decrease of acetylcholine (Ach) level. During PPN stimulation phase, the Glu concentration in VL was notably improved and the Ach concentration was mildly improved, but that of γ-GABA did not change.Conclusion Low frequency stimulation of PPN could rebalance the Glu and Ach level in VL nucleu of PD rats, which may be associated with its improving posture instability and gait difficulty.
