摘  要:目的探讨化疗对肝癌裸鼠原位移植瘤侵袭转移潜能的影响.方法选用稳定表达绿色荧光蛋白的Hep G2人肝癌细胞株构建肝癌裸鼠原位移植模型,成瘤后实验组给予10 mg/kg奥沙利铂腹腔注射化疗,每周1次,共3次.最后1次化疗后第7天,将2组裸鼠体内的残余肿瘤修剪成相同大小的瘤块再接种于新的裸鼠肝脏内.再接种后第6周,测量肿瘤大小,荧光显微镜下观察肿瘤的侵袭转移情况.结果对照组和化疗预处理组裸鼠肿瘤体积分别为(2 376.29±573.69)mm3和(1 637.23±338.68)mm3(P<0.05).对照组裸鼠肿瘤均未发生肝脏、腹腔和肺转移,化疗预处理组裸鼠肿瘤发生肝内转移率为83.3%(P=0.015),腹腔转移率为50%(P=0.182),肺转移率为50%(P=0.182).结论 Hep G2肝癌细胞在奥沙利铂化疗后生物学行为显著恶化.Objective To explore the effects of chemotherapy on the invasion and metastatic potential of orthotopic hepatocellular carcinoma nude mice model.Methods The orthotopic hepatocellular carcinoma nude mice model was establishedby using fluorescent Hep G2 cells. When the tumor reached approximately 1cm in length,oxaliplatin was administered intraperitoneally to randomly selected mice(experimental group,n=6) on days 12,19, and 26 after inoculation; micein the control group(n =6) received 0.2 m L of 0.9% sodium chloride intraperitoneally on those days. On day 7 after the final treatment, all mice were sacrificed by cervical dislocation.Tumor fragments of equal volume from each mouse of the oxaliplatin- treated and untreated control groups were reinoculated into the livers of each new recipient mice correspondently. 6 weeks after reinoculation,tumor volumes were assessed and the invasion and metastasis of tumors in each group were detected using a fluorescent microscope.Results Tumors in control group grew faster than those in experimental group [(2 376.29±573.69) mm3vs(1 637.23±338.68) mm3,P〈0.05]. No metastasis was found in the mice of control group. However,multiple metastasis were detected in the liver(0/6 vs 5/6,P=0.015), abdomen(0/6 vs 3/6,P=0.182) and lung(0/6vs 3/6,P=0.182) of the mice in the experimental group.Conclusion The biological features of Hep G2 cells aredeteriorated after oxaliplatin treatment.
