脑尔康对AD模型小鼠学习记忆及脑内AchE活性影响的拆方研究

摘  要:目的:对复方中药脑尔康进行拆方研究,筛选出治疗阿尔茨海默病(Alzheimer’s Disease,AD)模型小鼠药味精简,疗效较佳的组方。方法:70只小鼠随机分为7组,每组10只,采用腹腔注射氯化铝(AlCl3)溶液建立AD模型小鼠,空白组和模型组每天给予生理盐水0.5mL灌胃;其余各组每天分别给予相应药物14.3g/kg剂量灌胃,连续30d。观察各组小鼠学习记忆情况,并测定其右侧皮质、海马区AchE活性及其蛋白水平。结果:各用药组与模型组比较,全方及拆方1号学习记忆功能明显改善(P<0.05或P<0.01),海马、皮质区AchE活性均明显降低(P<0.01),其蛋白水平均明显升高(P<0.05或P<0.01);各拆方组分别与全方比较,拆方1号海马、皮质区AchE活性降低,其蛋白水平升高程度与全方无明显差异,拆方2,3,4号则均与全方有统计学差异(P<0.05或P<0.01);拆方2,3,4号与拆方1号比较,除拆方2号在提高海马区蛋白水平与拆方1号无明显差异外,其余各组均与拆方1号有统计学差异(P<0.05或P<0.01)。结论:拆方1号及全方改善AD模型小鼠学习记忆,抑制脑内AchE活性作用明显优于其他拆方组,两者疗效相当,且前者药味精简。Objective:Decompose the Naoerkang to screen one decomposed prescription which has better effect but has fewer ingredients on Alzheimer's Disease mice.Methods:70mice were randomly divided into 7groups,10 for each group,injected AlCl3 solution into stomach to made the AD model.Mice of the control and the model groups had been forced to drink saline 0.5mL per day;and mice of the other groups had been forced to drink the response medicine,14.3g/kg per day,for 30 days.observed the learning and memory ability and AchE activity in right cortex and hippocampus in each mouse.Result:Compared with the model group,the whole group and decomposed 1improved the learning and memory functions obviously(P〈0.05 or P〈0.01),decreased the AchE activity obviously(P〈0.01),increased the protein level evidently(P〈0.05 or P〈0.01)in hippocampus and cortex.All of its decomposed prescriptions compared with the whole group,there were no significant differences existed in decomposed prescription 1and the whole group,there were significant differences existed in decomposed prescription 2,3,4and the whole group(P〈0.05 or P〈0.01).Compared with the decomposed prescription 1,there were significant differences(P〈0.05 or P〈0.01)existed in decomposed prescription 2,3and 4except for increasing the protein level in hippocampus of decomposed prescription 2.Conclusions:decomposed prescription 1and Naoerkang groups both have better effect of improving the learning and memory functions and inhabiting the AchE activity than any other decomposed prescription,they have the same effect,but the former has fewer ingredients.
