摘  要:目的研究mi R-30c在前列腺癌组织的表达及其与前列腺癌的临床相关性。方法从mi RNA芯片检测结果中获得前列腺癌相关的特异表达mi R-30c分子,进一步运用q RT-PCR检测mi R-30c在前列腺癌细胞株及组织的表达情况,结合临床信息,分析mi R-30c表达与前列腺癌临床特征的相关性。结果前列腺癌mi RNA芯片结果表明,mi R-30c在肿瘤中表达下调显著(倍数变化=2.09倍,t=-3.65,P=0.016)。q RT-PCR检测mi R-30c的表达情况表明其在前列腺癌组织的表达显著低于良性组织(3.27±0.62 vs 6.85±0.33,t=7.53,P<0.001)。进一步分析mi R-30c表达与临床预后的关系,结果表明,mi R-30c的表达水平同Gleason评分(χ2=10.04,P<0.01)、病理分期(χ2=5.03,0.01<P<0.05)、转移(χ2=5.68,0.01<P<0.05)及生化复发(χ2=4.07,0.01<P<0.05)显著相关,即Gleason评分低组、病理分期低组、无转移和无生化复发的患者中的mi R-30c表达水平更高。经统计检验,它的表达水平同术前PSA水平(χ2=0.64,P>0.05)及切缘性质(χ2=0.64,P>0.05)无显著相关。结论 mi R-30c与前列腺癌临床相关,检测mi R-30c表达不仅可鉴别前列腺癌及良性组织,并可预测生化复发的概率及危险度。[ Objectives ] To study the expression of miR-30c in benign prostate hyperplasia (BPH) tissues and prostate cancer tissues, and to illustrate the clinical correlation between the miR-30c expression and prostate cancer. [ Methods ] miRNA microarray was used to detect the candidate microRNA genes for prostate cancer. The miR-30c expression in prostate cancer cell line and tissue was detected by qRT-PCR. The clinical prognosis value of miR- 30c was further analysed combining the clinical information. [ Results ] miR-30c was significantly down-regulated (fold change=-2.09, t = -3.65, P = 0.016) in primary prostate cancer tissue in miRNA microarray, qRT-PCR further confirmed that the expression of miR-30c was dramatically decreased in primary prostate cancer tissue compared with adjacent benign tissue (3.27±0.62 vs 6.85±0.33, t = 7.53, P 〈 0.001). The relation between miR-30c and clini- cal prognosis was analysed, which indicated the expression of miR-30c was correlated with Gleason scores (X2 = 10.04, P 〈 0.01), pathological stage (X2 = 5.03, 0.01〈 P 〈0.05), metastasis (X2 = 5.68, 0.01〈 P 〈0.05) and biochemi- cal recurrence (X2 = 4.07, 0.01〈 P 〈0.05). However, the level of miR-30c expression was irrelevant with the PSA level (X2 = 0.64, P 〉0.05) and the surgical margin status (X2 = 0.64, P 〉 0.05). [ Conclusions ] miR-30c related to the clinical features of prostate cancer, and the miR-30c expression level can be used to distinguish the prostate cancer and benign tissue, miR-30c may be a prognostic factor of biochemical recurrence.
