摘  要:原代肝细胞的分离培养技术已有50余年的历史,由于原代肝细胞有体内的代谢酶和相应辅助因子,能维持肝脏的代谢功能,是研究药物代谢和致癌作用理想的体外模型。随着近10年来新技术的不断发展,尤其是三维培养技术的建立以及毒理基因组学的应用,使得体外原代肝细胞模型能用于对化合物潜在致癌性风险的预测,成为临床前药物致癌性实验的一种替代方法。本文对原代肝细胞分离、培养技术及其作为致癌物预测体外模型应用的研究进展进行综述和展望。During the past several decades, primary hepatocyte isolation and culture technique has been extensively utilized. As it can reserve the metabolism function, enzymes and corresponding cofactor activity of liver, primary hep- atocytes serves as an ideal in vitro model for assessing drug metabolism and the mechanism of carcinogens. With the development of new techniques during the last decade, especially the three-dimensional culture systems and toxicogenomies,the primary hepatocyte in vitro models can be used to predict the potential carcinogenicity of carcinogenic substances,which has become a new alternative approach for non-clinical carcinogenicity study of drugs. This review focuses on the progress of primary hepatocyte isolation and culture techniques, as well as its future application as in vitro models to predict potential carcinogens.
