摘 要:目的观察N-甲基-D-天门冬氨酸受体阻滞剂MK-801对6-羟基多巴胺(6-OHDA)诱导的帕金森病(PD)模型大鼠黑质-纹状体通路的作用。方法将40只大鼠随机分为正常对照组、6-OHDA模型组、MK-801低剂量(0.1mg/kg)处理组和MK-801高剂量(0.2mg/kg)处理组。采用6-OHDA单侧损毁大鼠黑质制备PD模型。应用免疫组织化学法检测各组大鼠黑质内酪氨酸羟化酶(TH)免疫阳性神经元数目,高效液相色谱法检测纹状体内多巴胺(DA)的含量。结果与正常对照组相比,6-OHDA模型组大鼠6-OHDA损毁侧中脑黑质TH免疫阳性神经元数目、纹状体内DA含量显著降低(F=478.02、34.86,q=51.25、14.09,P〈0.05)。与6-OHDA模型组相比,MK-801低剂量处理组和MK-801高剂量处理组大鼠黑质TH免疫阳性神经元数目、纹状体内DA含量均显著增加(q=4.27~14.96,P〈0.05)。结论 MK-801对PD模型大鼠黑质-纹状体通路有明显的保护作用。Objective To explore the protective effect of MK-801 on nigrostriatal projection system in 6-hydroxydopamine(6-OHDA)-induced Parkinson disease(PD)models. Methods Forty rats were randomized to control group,6-OHDAtreated group,0.1mg/kg MK-801 pretreated group and 0.2mg/kg MK-801 pretreated group.6-OHDA was used to create PD rat models.Applying immunohistochemical method and high performance liquid chromatography to detect the number of tyrosine hydroxylase(TH)-positive neurons in the substantia nigra(SN)and dopamine contents in the striatum of rats,respectively. Results The number of TH-positive neurons in the SN and dopamine contents in the striatum decreased significantly in 6-OHDAtreated rats compared with the controls(F=478.02,34.86;q=51.25,14.09;P0.05).Compared with 6-OHDA-treated group,the numbers of TH-positive neurons and striatal DA contents in low-dose-treated and high-dose-treated groups obviously increased(q= 4.27-14.96,P0.05). Conclusion MK-801 shows a notable protective effect on nigrostriatal path action in rat models of 6-OHDA-induced PD.
