炎性因子在大鼠牙周炎合并血管钙化模型中的表达

摘　　要：目的通过比较大鼠牙周炎模型、血管钙化模型及联合模型血清中牙周炎性因子C-反应蛋白（CRP）、白细胞介素6（IL-6）、肿瘤坏死因子α（TNF-α）的含量,初步探讨牙周炎和血管钙化之间的关系。方法SPF级成年雄性Wistar大鼠32只,随机分为正常对照组（C组）8只,牙周炎组（CP组）9只,血管钙化组（VDN组）9只,血管钙化＋牙周炎组（CP＋VDN组）6只,4组接受相应的建模处理,测体质量,8周后处死,采用酶联免疫吸附法检测各组血清中CRP、IL-6、TNF-α的含量。结果 CP＋VDN组和CP组血清CRP、IL-6、TNF-α的水平显著高于C组和VDN组（F=56.02~91.21,q=1.80~11.80,P〈0.05）,VDN组以上指标较C组显著增高（q=3.96~10.24,P〈0.05）。结论 CRP、IL-6、TNF-α与牙周炎和血管钙化关系密切,当两者同时存在时,血清炎性因子的含量可能叠加。Objective To compare serum content of C-reactive protein（CRP）,interleukin-6（IL-6）and tumor necrosis factor-α（TNF-α）in periodontal model,vascular calcification model,and compound model of rat,and preliminarily explore the correlation between periodontal and vascular calcification. Methods Thirty-two adult male Wistar rats were randomly divided into control group（C group,n=8）,periodontitis group（CP,n=9）,vascular calcification group（VDN,n=9）and compound group（CP＋VDN,n=6）.The rats in each group were given corresponding managements to create an animal model,the body weight was measured.All the rats were sacrificed eight weeks later,and serum CRP,IL-6and TNF-αwere detected by ELISA method.Results The serum levels of CRP,IL-6,and TNF-αin CP＋VDN and CP groups were notably higher than that in VDN and C groups（F=56.02-91.21,q=1.80-11.80,P0.05）,and that in VDN group was higher than C group（q=3.96-10.24,P0.05）. Conclusion CRP,IL-6,and TNF-αare closely associated with periodontitis and vascular calcification.When both of the disease exist at the same time,the amount of inflammatory cytokines in serum may be superimposed.
