摘 要:目的探讨miRNA-26a、miRNA-143、miRNA-195在家族性乳癌组织和血清中表达变化,以评价其对家族性乳癌早期诊断的潜在价值。方法收集家族性乳癌、散发性乳癌病人的癌组织、癌旁正常组织及血清标本,并收集健康查体女性血清标本作为对照。提取血清及组织标本中的总RNA,进行逆转录和实时荧光定量PCR检测miRNA-26a、miRNA-143、miRNA-195在组织和外周血表达情况。结果乳癌组织中miRNA-26a、miRNA-143、miRNA-195的表达水平均明显低于癌旁对照组织(t=2.033~2.671,P〈0.05)。家族性乳癌组织中miRNA-26a、miRNA-143表达水平低于散发性乳癌组织(Z=1.978、2.315,P〈0.05)。乳癌病人血清miRNA-26a、miRNA-143、miRNA-195表达水平均明显低于对照组(Z=2.211~5.922,P〈0.05);家族性乳癌病人血清中miRNA-143、miRNA-195表达水平低于散发性乳癌组(Z=3.103、2.478,P〈0.05)。miRNA-143在组织与血清中的表达水平呈正相关(r=0.545,P〈0.05)。结论 miRNA-26a、miRNA-143、miRNA-195在乳癌病人癌组织及血清中表达水平下调,可能成为乳癌筛查和诊断的潜在生物学指标;miRNA-143在家族性乳癌病人癌组织及血清中表达水平明显下调,可能成为家族性乳癌诊断的生物学指标。Objective To investigate the expressions of miRNA-26 a,miRNA-143,and miRNA-195 in serum and tissue of patients with familial breast cancer(BC) and assess their potential value in the early diagnosis of this type of malignancy.Methods Cancer and paraneoplastic tissue and serum specimens of patients with familial and sporadic BC were collected,and healthy serum samples collected from women who came for medical examination were served as controls.Total RNA were extracted from all the serum nnd tissue samples,the expressions of three miRNAs(miRNA-26 a,miRNA-143,miRNA-195) were quantified by using real-time PCR technique.Results The expressions of miRNA-26 a,miRNA-143 and miRNA-195 in cancer tissues were lower than that in paraneoplastic specimens(t=2.033-2.671,P〈0.05),and that in familial BC were lower than that in sporadic BC(Z=1.978.2.315;P〈0.05).The expressions of serum miRNA-26 a,miRNA-143 and miRNA-195 in the BC patients were lower than that in the normal controls(Z=2.211-5.922,P〈0.05),that familial BC were lower than that in sporadic BC(Z=3.103,2.478;P〈0.05).A positive correlation was noted between tissue miRNA-143 expression and serum miRNA-143(r=0.545,P〈0.05).Conclusion Down regulation of serum and tissue miRNA-26 a,miRNA-143 and miRNA-195 in breast cancaer patients is likely to become a potential biological parameter for screening and diagnosis of breast cancer,and significant down regulation of tissue and serum miRNA-143 in familial breast cancer can probably become a biological parameter in the diagnosis of this familial malignancy.
