胃癌组织MUC1表达及其与幽门螺杆菌感染关系

摘　　要：目的研究幽门螺杆菌(HP)对胃癌病人黏蛋白1(MUC1)表达的影响,探讨HP感染影响胃癌浸润转移的机制。方法建立HP感染胃癌AGS细胞系模型,Heochest33258染色观察HP感染的AGS细胞形态学变化,Western blot方法检测HP感染对AGS细胞MUC1表达的影响;免疫组织化学方法检测96例胃癌组织中MUC1的表达。结果 Heochest33258染色显示,HP感染后AGS细胞数目减少,细胞核出现浓缩、荧光染料浓聚等典型的凋亡现象。Western blot结果显示,HP感染后AGS细胞MUC1表达上调;MUC1在HP感染的胃癌组织中表达率为67%(33/49),在HP阴性的胃癌组织中表达率为45%(21/47),差异有统计学意义(χ2=5.008,P<0.05)。结论 HP感染影响胃癌组织MUC1的表达,HP感染引起MUC1蛋白表达上调可能促进胃癌转移。Objective To study the influence of helicobacter pylori（HP）infection on the expression of MUC1 in gastric cancer,and explore the effect of HP on infiltration and metastasis of this malignancy. Methods A HP-infected AGS-cell-line model was created.Heochest33258 staining done to observe morphologic variations of AGS infected with HP,the effect of HP infection on the expression of MUC1 in AGS was detected using western blot,and the expression of MUC1 in 96patients with gastric cancer was detected using immunohistochemical method. Results Heochest33258 staining showed that after HP infection,the number of AGS cells decreased,and typical manifestations of apoptosis-nucleus and fluorochrome concentrated-appeared.Western blot indicated after HP infection,the expression of MUC1 increased.The expression rate of MUC1 was 67%（33/49）in HP-positive gastric cancer versus 45%（21/47）in HP-negative,the difference being significant（χ^2=5.008,P〈0.05）. Conclusion Helicobacter pylori infection affects MUC1 expression in tissue of gastric cancer,the up-regulation of MUC1 expression caused by Helicobacter pylori is likely to be one of the mechanisms to facilitate metastasis for this malignancy.
