摘  要:以人弥漫大B细胞淋巴瘤(diffuse large B-cell lymphoma,DLBCL)活化B细胞(activated B-cell,ABC)型细胞株OCI-Ly3为研究对象,探索ABC型的生物学特性及肿瘤相关分子的表达,以及小鼠成瘤条件.对不同条件下培养的OCI-Ly3细胞进行对比分析,观察细胞生长形态及生物学特性;分析肿瘤相关抗原在OCI-Ly3细胞的表达情况;采用皮下和腹腔接种,在SCID小鼠中建立移植瘤动物模型.流式分析显示,体外培养的OCI-Ly3细胞对c-Met、Ki-67和survivin等细胞增殖和凋亡相关标记物具有较高的表达;SCID小鼠皮下接种107 OCILy3细胞,成瘤率87.5%;腹腔接种2×107细胞,成瘤率75%;肿瘤组织符合人DLBCL的组织学特征,且分布弥散,具有较高的侵袭性.成功构建了人ABC型DLBCL的小鼠模型,并对OCI-Ly3细胞的体外培养特性和与肿瘤发生相关的免疫标记物进行了较为系统的检测,为DLBCL的进一步研究提供了参考和工具.Objective Diffuse large B-cell lymphoma (DLBCL) is an aggressive malignancy of human B- cell lymphomas, with poor efficacy of clinical treatment and prognosis. We used an activated B-cell-like subtype cell line, OCI-Ly3, to investigate immunological characteristics and establish an animal model for human DLB- CL. Methods OCI-Ly3 cells were cultured under different culture conditions, and the morphology characteris- tics of cells were inspected. Expression of tumor-related markers was analyzed. Tumor was established in SCID mice via subcutaneous (s. c. ) or intraperitoneal (i. p. ) routes. Results Median expression of B cell-related markers and high expression of proliferation/apoptotic-related markers were observed on OCI-Ly3 cells. Tumor were successfully established by s.c. (87.50../00, 194-1.2 d) or i.p. (75%) injection of OCI-Ly3 cells in SCID mice. Conclusion Our study will provide valuable reference and animal model for further research of human DL- BCL.
