摘  要:目的研究原发性肾小球疾病患者肾组织中TLR2和TLR4mRNA和蛋白的表达特点,并探讨其与炎性因子的相关性。方法2010年1月到2014年4月我院共收治原发性肾小球疾病住院患者24例(标记为观察组),另选择18例怀疑肾病但经肾活检没有患病(标记为对照组)的患者,全自动生化分析仪检测血清肾功能,试剂盒检测其炎性因子及TLR2和TLR4含量,免疫组化、WestemBlot以及半定量PCR,测定肾组织TLR2和TLR4mRNA及蛋白表达水平。结果观察组患者肾功能指标血尿素氮、血肌酐、血尿酸,炎性因子IL-8、IL-10、IL-23、TNF-α,TLR2和TLR4明显高于对照组(P〈0.05)。结论原发性肾小球疾病除肾功能明显异常外还有明显的炎症反应,其机制可能是TLR2和TLR4通过激活相关炎性因子表达有关。Objective To study TLR2 and TLR4 mRNA and protein expression in the renal tissue of patients with primary glomerular disease, and to explore the role of TLR2 and TLR4 in this diseases. Methods From January 2010 to April 2014 were treated in primary glomerular disease patients 24 cases (labeled B group), randomly selected 10 cases of kidney disease but do not speak up surgery in patients with other diseases (marked for group A), automatic biochemical analyzer renal function, Western Blot and semi-quantitative PCR, determination of renal tissue TLR2 and TLR4 mRNA expression levels. Results The blood urea nitrogen, serum creatinine, serum uric acid, inflammatory factor IL-8, IL-10, IL-23, TNF-α, TLR4 and TLR2 were significantly higher in the observation group than in the control group (P〈0.05).Conclusion Primary glomerular diseases are significant inflammatory response, the mechanism may be related by activating TLR2 and TLR4 expression of inflammatory cytokines caused.
