摘  要:目的研究重组高效复合干扰素(rSIFN-co)对肺腺癌细胞株A549增殖和凋亡的影响。方法筛选rSIFN-co对A549细胞的有效浓度,及与顺铂联合用药的最佳浓度和作用时间。分为rSIFN-co(5μg/mL)组、传统重组集成干扰素α(干复津,5μg/mL)组、rSIFN-co(5μg/mL)+顺铂(2μg/mL)组、干复津(5μg/mL)+顺铂(2μg/mL)组、顺铂(2μg/mL)组和RPMI-1640液空白组,分别干预A549细胞。MTT法检测各组7d活细胞数并作生存曲线,流式细胞仪检测48h各组细胞凋亡率,免疫荧光法检测48h各组凋亡相关蛋白Fas、Bcl-2表达。结果 rSIFN-co的有效浓度为1-64μg/mL,顺铂的最低有效浓度为2μg/mL,与2μg/mL顺铂联合用药时rSIFN-co的最佳浓度为5μg/mL,最佳作用时间为48h。生存曲线示rSIFN-co对A549的抑制作用强于干复津,rSIFN-co+顺铂的抑制作用强于干复津+顺铂。rSIFN-co可诱导A549细胞凋亡,rSIFN-co组凋亡率高于干复津组(P=0.000),rSIFN-co+顺铂组凋亡率高于rSIFN-co组(P=0.004)及顺铂组(P=0.023)。rSIFN-co能增强A549中Fas的表达,并抑制Bcl-2的表达,其中rSIFN-co组Bcl-2表达低于干复津组(P〈0.05)。结论 rSIFN-co能抑制A549增殖,且作用强于干复津,与顺铂联合应用的抑制作用增强,其对A549的抑制作用与调节凋亡相关基因表达有关。Objective To investigate the effect of recombinant super-compound interferon(rSIFN-co)on the proliferation and apoptosis of pulmonary adenocarcinoma cell line A549.Methods Screening tests were conducted to determine the concentrations of rSIFN-co that have a significant impact on A549 and the optimal concentration and duration for the test of rSIFN-co combined with Cisplatin.A549 cells were treated with rSIFN-co,Infergen,rSIFNco+Cisplatin,Infergen+Cisplatin,and Cisplatin,respectively,and compared with those cultured in normal medium.The viable A549 cells from Day 1to Day 7were detected by MTT assay.Cell apoptosis was detected by flow cytometry(FCM).Apoptosis-associated proteins,Fas and Bcl-2 were detected by immunofluoroscence at48 h.Results Effective concentrations of rSIFN-co ranged from 1to 64μg/mL,and a minimal of 2μg/mL Cisplatin was needed.The optimal test condition was set at 5μg/mL rSIFN-co combined with 2μg/mL Cisplatin for a duration of 48 h.rSIFN-co demonstrated a stronger inhibiting effect on cell proliferation than Infergen.The inhibiting efficiency of rSIFN-co+Cisplatin was also stronger than that of Infergen+Cisplatin.Apoptosis of A549 cells induced by rSIFN-co was also more significant than that of Infergen(P=0.000).Cells treated with rSIFNco+ Cisplatin has a higher apoptosis rate than those treated with rSIFN-co(P=0.004)or Cisplatin(P=0.023).rSIFN-co increased the expression of Fas and decreased the expression of Bcl-2.Cells treated with rSIFN-co showed lower fluoroscence intensity of Bcl-2than those treated with Infergen(P 〈0.05).Conclusion rSIFN-co inhibits the proliferation of A549 and its effect is stronger than that of Infergen.Cisplatin can further enhance the inhibiting effect of rSIFN-co.The inhibiting efficiency may be associated with the expression of apoptosis-related genes.
