摘 要:目的研究烟碱对人牙周膜干细胞(periodontal ligament stem cell,PDLSC)成骨能力的影响。方法运用流式细胞仪检测有限稀释法培养的PDLSC的表面抗原标志物,采用细胞增殖检测试剂盒检测分别经10-3mol/L、10-5mol/L、10-7mol/L浓度烟碱刺激后PDLSC增殖能力变化。运用碱性磷酸酶定性染色和实时定量PCR检测不同浓度烟碱刺激及烟碱联合拮抗剂α-银环蛇毒素(α-bungarotoxin,α-BTX)刺激后,PDLSC成骨能力的改变。结果经过有限稀释法所得的细胞阳性表达间充质干细胞的标志物,不同浓度的烟碱可抑制PDLSC的增殖及成骨能力,加入拮抗剂α-BTX后,烟碱抑制PDLSC成骨的作用降低。结论烟碱能够抑制PDLSC的成骨能力。Objective To study the effect of nicotine on proliferation and osteogenic ability of human periodontal liga-ment stem cells ( hPDLSCs) .Methods Flowcytometer was applied to detect the surface antigen markers of hPDLSCs. WST-1 was used to detect the proliferation ability of hPDLSCs which was stimulated by different concentration of nicotine, with 10 -3 mol/L, 10 -5 mol/L, 10-7 mol/L respectively.ALP and RT-PCR were used to detect the change of osteogenic ability of hPDLSCs stimulated by different concentration of nicotine when givenα-BTX competitive antagonist of nicotine. Results Normal mesenchymal tissues surface markers were detected in hPDLSCs.Different concentrations of nicotine could suppress the cell proliferation and osteogenic ability.After givenα-BTX, the suppressive impact of nicotine could be partially reversed.Conclusion Nicotine could suppress the proliferation and osteogenic ability of hPDLSCs.
