摘 要:目的研究细胞周期蛋白D2(cyclin D2)对HBV复制的影响并探讨其作用的分子机制。方法运用qRT-PCR分析细胞周期蛋白家族成员在HBV复制细胞系中的表达水平;qRT-PCR、Western blot检测HBV复制对cyclin D2表达的影响;Western blot验证cyclin D2沉默效果,Real-time PCR,Southern blot验证沉默cyclin D2对HBV复制中间体表达的影响及ELISA验证cyclin D2对HBs Ag、HBe Ag分泌的影响;并检测cyclin D2过表达对HBV复制的影响。结果在细胞周期蛋白中,HBV复制对cyclin D2表达的影响最为显著,HBV感染明显促进cyclin D2 mRNA和蛋白水平的升高,下调cyclin D2显著抑制HBV的复制中间体的表达,HBs Ag和HBe Ag的分泌,过表达cyclin D2明显促进HBV的复制。结论 HBV复制可以促进细胞内cyclin D2的表达,cyclin D2有利于HBV的复制。这一发现为降低HBV的感染率,探索治疗乙型肝炎新的靶点提供了新的策略。Objective and explore the cyclin D2 regulati To investigate the role of cyclin D2 in hepatitis B virus (HBV) replication on mechanism in HBV replication. Methods The mRNA levels of cyclin family members were analyzed by RT-PCR. The effect of cyclin D2 on the expression of HBV replicative intermediates was measured by real-time PCR and Southern blotting, and the effect of cyclin I)2 on the secretion of HBsAg and HBeAg was analyzed by ELISA. The effect of cyclin D2 over-expression on HBV replication was analyzed. Results Among the cyclin family members, HBV replication had the most significantly impact on cyclin D2 expression, and HBV replication induced cyclin D2 replication. Cyclin D2 knockdown inhibited the expression of HBV replicative intermediates and the secretion of HBsAg and HBeAg, and cyclin D2 over- expression promoted HBV replication. Conclusion HBV replication promotes cyelin D2 expression, and cyclin D2 is conducive to HBV replication. This discovery can provide a novel strategy for reducing HBV infection and exploring new hepatitis B therapeutic target.
