摘 要:[目的]研究参芪泄浊饮对腺嘌呤致大鼠慢性肾衰肾组织基质金属蛋白酶1(MMP-1)、基质金属蛋白酶抑制剂1(TIMP-1)基因表达的影响。[方法]SPF级雄性SD大鼠54只,采用Yokozawa法复制慢性肾衰大鼠模型,随机分为参芪泄浊饮高剂量组、参芪泄浊饮常规剂量组、氯沙坦组、模型组,分别予高剂量组、常规剂量组、氯沙坦组以参芪泄浊饮水煎液20m L/kg·d、10m L/kg·d、氯沙坦组9mg/kg·d,模型组与空白组大鼠均予纯净水10m L/kg·d,连续灌胃干预8周,ELISA法测量血肌酐(Scr)、尿素氮(BUN)、丙氨酸转氨酶(ALT);荧光定量PCR方法检测肾组织中MMP-1、TIMP-1。[结果]1Scr、BUN各组大鼠较空白组均有升高(P<0.05),氯沙坦组明显低于模型组(P<0.05);Scr高剂量组大鼠明显低于常规剂量组、氯沙坦组及模型组大鼠(P<0.05),常规剂量组与氯沙坦组亦低于模型组(P<0.05);BUN变化趋势与Scr相同,高剂量组降低不明显(P>0.05)。2MMP-1 m RNA各组均高于空白组(P<0.05),高剂量组显著高于常规剂量组、氯沙坦组及模型组(P<0.05),常规剂量组与氯沙坦组也显著高于模型组(P<0.05);TIMP-1 m RNA各组均高于空白组(P<0.05),高剂量组明显低于常规剂量组、氯沙坦组及模型组(P<0.05),常规剂量组与氯沙坦组明显低于模型组(P<0.05)。[结论]参芪泄浊饮可通过促进MMP-1 m RNA及抑制TIMP-1 m RNA表达,使细胞外基质(extracellular matrix,ECM)的降解增加,堆积减少,进而减缓CRF的进一步发展,保护肾脏。[Objective]To investigate the effects of Shenqi Xiezhuo decoction on matrix metalloproteinase- 1(MMP-1)and tissue inhibitor of matrix metalloproteinase-1(TIMP-1) expression in adenine-induced renal interstitial fibrosis rats.[Methods]SPF male SD rats were 54,the renal failure model of the remaining rats were established by Yokozawa method.All rats were randomly devided into 5 groups: high-dose group,regular-dose group,losartan group,model group and control group.Model group and the control group of rats herein are pure water.After 8 weeks,all rats were collected to analyz by serum biochemical tests and RT-PCR.Results:(1)The level of serum crcatinine of high-dose group was significantly lower than the levels of regular-dose group or losartan group,and there was no significant difference between the latter two groups.(2)The expression of MMP-1 mRNA of high-dose group was more than the regular-dose group or losartan group,and there was no significant difference between the latter two groups.The expression of TIMP-1 mRNA of high-dose group was less than the regular-dose group or losartan group,and there was no significant difference between the latter two groups.(3)There was no difference of the level of ALT between control group and the others.Conclusions: Shenqi Xiezhuo decoction may alleviate deterioration of the renal function by regulating MMP-1 and TIMP-1 balance.
