摘　　要：目的探讨基质交感分子1(stromal interaction molecule one,STIM1)在急性坏死性胰腺炎(acute necrotizing pancreatitis,ANP)大鼠胰腺组织的表达变化。方法 60只SD大鼠分为假手术组和模型组;假手术组仅开腹翻动胰腺,模型组逆行胰胆管注射2%牛磺胆酸钠诱导大鼠ANP模型。分别在3、6、12、24、48 h时处死大鼠,取胰腺组织进行病理学评分,检测血清淀粉酶浓度;应用荧光定量PCR检测胰腺组织STIM1 mRNA表达,应用蛋白印迹法检测胰腺组织STIM1蛋白表达水平。结果模型组大鼠胰腺病理评分、血清淀粉酶高于假手术组(P<0.05),胰腺组织STIM1 mRNA在24、48 h时低于假手术组(P<0.05);胰腺STIM1蛋白在24 h时低于假手术组(P<0.01)。结论实验性ANP大鼠胰腺STIM1 mRNA和蛋白表达水平在发病后24 h降低,可能与AP的钙稳态失衡有关。Objective To investigate the change of expression of stromal interaction molecule one（ STIM1） in acute necrotizing pancreatitis（ ANP） rats. Methods Sixty male SD rats were randomly divided into two groups： sham operation group and model group. Rats in model group received sham operation and pancreas flipping. Rats in model group was induced by injecting 2% sodium taurocholate retrogradely into biliopancreatic duct. All rats were sacrificed at 3rd,6th,12 th,24th,48 th hour after ANP induction. Pathological changes in pancreatic tissue were observed and scored.Serum amylase（ AMS） was determined by ELISA. The expressions of STIM1 mRNA and protein in pancreas were evaluated by quantitative PCR and Western blotting. Results The pathologic scores,AMS of model group were significantly higher than those of sham operation group. Serum AMS levels of group A were higher than those of group C（ P〈0. 05）.Compare with sham operation group,the expression level of STIM1 mRNA in model group was lower than that in the sham operation group at 24 th and 48 th hour. At the same time,the expression of STIM1 protein was lower than that in the sham operation group at 24 th hour. Conclusion The expressions of STIM1 mRNA and protein in pancreatic tissue are decreased in the ANP rats. STIM1 maybe play a role in maintaining the calcium homeostasis in pancreatic acinar cell.
