摘  要:目的:建立柱前衍生化HPLC检测不同基原珍珠粉及贝壳粉中7种主要氨基酸的方法。方法:以异硫氰酸苯酯为柱前衍生化试剂,采用C18色谱柱,流动相A:0.1mol/L醋酸钠溶液(用醋酸调pH值至6.5),流动相B:乙腈,梯度洗脱,检测波长254nm。结果:氨基酸衍生溶液在24h内保持稳定,7种氨基酸线性相关系数均大于0.9995,加样回收率在93.16%~103.31%之间。结论:本方法分离效果好,精密度和重复性良好,可用于珍珠粉及贝壳粉中7种水解氨基酸含量的测定。Objective: To establish an analysis method for determination of amino acids in pearl powder and conch powder from different origins by HPLC with pre-column derivation. Methods: Hydrolytic amino acids were extracted by acid hydrolysis method,then derivatized with phenyl isothiocyanate( PITC). And the samples were analyzed on a C18 column with gradient elution of 0. 1 mol / L sodium acetate solution( adjusted to p H 6. 5)( A) and acetonitrile( B) at the flow rate of 1. 0 m L / min. The detected wavelength was 254 nm. Results: Amino acids derivative solution remained stable in 24 hours. The response was linear for seven amino acids with a correlation coefficient r 0. 9995. The average recoveries were 93. 16% ~ 103. 31%. Conclusion: The method is sensitive with good accuracy and repeatability,which is useful for the determination of amino acids in pearl powder and conch powder from different origins.
