染色体1p/19q缺失与P53、IDH1突变蛋白在少突胶质细胞肿瘤中的表达及相关性

摘　　要：目的研究少突胶质细胞肿瘤染色体1p/19q杂合性缺失与人异柠檬酸脱氢酶1-R132H(isocitrate dehydrogenas 1-R132H,IDH1-R132H)、P53突变蛋白表达的相关性。方法选取病理学诊断为少突胶质细胞肿瘤标本,用荧光原位杂交方法检测染色体1p/19q杂合性缺失情况,用免疫组织化学法检测IDH1-R132H、P53突变蛋白表达情况,分析染色体1p/19q缺失与IDH1、P53突变蛋白表达之间的相关性。结果 76例少突胶质细胞肿瘤1p/19q杂合性缺失率为73.7%,均为联合缺失。少突胶质细胞瘤1p/19q杂合性缺失率比间变型少突胶质细胞瘤高,但差异无统计学意义。56例1p/19q杂合性缺失的少突胶质细胞肿瘤均为IDH1-R132H突变蛋白表达阳性,其中54例均为P53突变蛋白表达阴性。IDH1、P53突变蛋白表达水平与染色体1p/19q杂合性缺失具有相关性(P<0.05)。结论少突胶质细胞肿瘤染色体1p/19q杂合性缺失与人IDH1-R132H突变蛋白表达、P53突变蛋白阴性表达呈正相关。Objective To study the correlation between chromosome 1p /19 q deletion and expressions of R132 H mutant isocitrate dehydrogenas 1（ IDH1）,P53 in oligodendroglial tumors.Methods Seventy-six cases of oligodendroglial tumors were collected. Expressions of R132 H mutant IDH1 protein and P53 were detected by immunohistochemistry,1p /19 q deletion was detected with fluorescence in situ hybridization（ FISH）,and correlation between chromosome1 p /19 q deletion and expression of R132 H mutant IDH1 status was analyzed. Results FISH studies showed that deletion of chromosome 1p /19 q was found in 56 cases,there as no significant difference in deletion rate of chromosome 1p /19 q between oligodendrogliomas and anaplastic oligodendrogliomas. Among the 56 cases,negative expression of mutant P53 protein was found in 54 cases,positive expression of IDH1-R132 H was found in 56 cases. There was a statistical correlation between the expressions of R132 H mutant IDH1 protein,P53 and the 1p /19q combined deletion in oligodendroglial tumors（ P〈0. 05）. Conclusion 132 H mutant IDH1 protein and P53 may be the potential molecular marker forecasting 1p /19 q deletion in oligodendroglial tumors.
