摘 要:目的研究叶酸受体(FR)α在垂体腺瘤中的表达及意义,旨为垂体腺瘤的靶向诊治提供理论依据。方法采用RT—PCR、Westernblot及免疫组织化学法分别检测垂体腺瘤和正常垂体组织中FR仪的mRNA和蛋白表达水平,并研究FRα的表达和部分临床指标的关系。结果RT-PCR方法显示在无功能性(NF)垂体腺瘤中FRα的mRNA表达量显著高于有内分泌功能的垂体腺瘤及正常垂体组织(P〈0.05);Westernblot结果显示FRⅡ在NF垂体腺瘤中明显高表达(P〈0.05),而在有内分泌功能的垂体腺瘤及正常垂体组织中几乎不表达;免疫组织化学法显示FRct仅在NF垂体腺瘤中表达,表达阳性率为93.75%(30/32)。此外,FR仅在侵袭性垂体腺瘤中的表达显著高于非侵袭性垂体腺瘤(P〈0.05),但其表达量与患者的性别、年龄、肿瘤直径、复发性肿瘤无明显相关性(P〉0.05)。结论FRα在NF垂体腺瘤中的表达明显高于有内分泌功能的垂体腺瘤及正常垂体组织,且其可能与垂体腺瘤的侵袭性有关联,这对NF垂体腺瘤将来的分子靶向诊断和治疗提供一定的研究基础。Objective To study the expression and significance of folate receptor (FR)α in pituitary adenomas in order to provide a theoretical basis for targeting the diagnosis and treatment of pituitary adenomas. Methods RT-PCR, Western blot and immunohistochemistry were used to detect mRNA and protein expression levels of FRα in pituitary adenomas and normal pituitary tissue, and the relationship between the FRα expression and part of the clinical indicators were studied. Results Real-time quantitative polymerase chain reaction (RT-PCR) showed that the mRNA expression level of FRα in nonfunctioning (NF) pituitary adenomas was significantly higher than that of pituitary adenomas with endocrine function and normal pituitary tissue ( P 〈 0.05 ). The result of Western blot showed that FRα expressed significantly high in NF pituitary adenomas (P 〈 0.05 ), while FRα hardly expressed in pituitary adenomas with endocrine function and normal pituitary tissue. Immunohistochemistry assay showed that FRα only expressed in NF pituitary adenomas, and the positive rate of expression was 93.75% (30/32). Furthermore, the expression of FRet in invasive pituitary adenomas was significantly higher than that in noninvasive ones (P 〈 0.05 ) , but there were no significant correlations in the expression levels of the sex, age, tumor diameter and recurrent tumor of the patients (P 〉 0.05 ). Conclusions The expression of FRα in NF pituitary adenomas was significantly higher than that in pituitary adenomas with endocrine function and normal pituitary tissue, and it might be associated with the invasiveness of pituitary adenomas. This provided a certain research foundation for NF adenomas in future molecular targeted diagnosis and treatment.
