COX-2和MMP-2在结直肠癌浸润转移中的表达及意义

摘　　要：目的:本课题通过检测COX-2与MMP-2在结直肠癌组织和正常结肠组织中的表达情况,探讨它们与结直肠癌病例的各项临床病理参数的关系及相关性,了解COX-2和MMP-2在结直肠癌浸润和转移中的临床病理意义。方法:本文收集了结直肠癌根治术后病理存档的石蜡标本117例及正常结直肠黏膜组织15例。应用免疫组织化学方法检测COX-2和MMP-2表达水平并分析其在结直肠癌组织中的表达及其临床病理学意义。结果:COX-2及MMP-2在结直肠癌组织中的阳性表达明显高于正常结直肠黏膜组织,两者差异具有统计学意义(P<0.05)。COX-2及MMP-2在结直肠癌组织中的表达与肿瘤浸润深度、分化程度、淋巴结转移情况、TNM分期间差异均有统计学意义(均P<0.05)。结论:COX-2、MMP-2在结直肠癌组织中的表达明显高于正常结直肠组织,且肿瘤中的表达有着正相关性有着协同效应,表明COX-2、MMP-2通过一定的机制相互作用,共同参与结直肠癌的浸润转移,这可能对结直肠癌的诊治有一定的帮助。Objective： In the thesis,the author test the expression of cyclooxygenase-2（ COX-2）and matrix metalloprote inase-2（ MMP-2） in colorectal cancer tissues and normal colon tissues,and detect the correlation of colorectal cancer cases in the clinic pathological parameters,to further understand the clinic pathological sense of the COX-2 and MMP-2 in the invasion and metastasis of colorectal cancer. Methods： 117 paraffin specimens in the case of colorectal cancer after operation and 15 in the cases of normal colorectal mucosa resection were collected. Immunohistochemical method was used to detect the level of expression of COX-2 and MMP-2,and analysis the difference of expression of COX-2 and MMP-2 in colorectal cancer tissue and normal colorectal mucosa tissues. Results： the positive expression of COX-2 and MMP-2 in colorectal cancer tissues was significantly higher than that in normal colorectal mucosa,the difference was statistically significant（ P 〈0. 05）. There was statistically significant difference for the expression of COX-2 and MMP-2 in colorectal cancer tissues with tumor cell invasion,tumor differentiation,lymph node metastasis,TNM stage,（ P 〈0. 05）. While no significant differences with gender,age,tumor location,tumor diameter,liver metastasis,and survival status（ P 〈0. 05）. The expression of COX-2 and MMP-2 had significant positive correlation. Conclusion： the expression of COX-2,MMP-2 in colorectal cancer tissues was higher than that in normal colorectal tissues,and the expression of COX-2 and MMP-2 in tumor tissues have a positive correlation with synergistic effect,which prove that COX-2、MMP-2 plays a role in tumor occurrence,development,and they can be used for judging tumor occurrence,and can be useful for the prognosis,diagnosis and treatment of tumor.
