生物酶解法提取匙羹藤总皂苷工艺的研究

摘　　要：目的通过正交实验优选纤维素酶法提取匙羹藤茎总皂苷的最佳提取工艺。方法匙羹藤总皂苷在5%香草醛-冰醋酸和高氯酸的混合体系中显色,于500 nm处有最大吸收,用比色法测总皂苷含量,以总皂苷的产量为考察指标。采用酶解-乙醇浸提,正丁醇萃取,匙羹藤总皂苷提取率作为评价指标,通过单因素试验和正交试验,确定提取最佳工艺。结果酶解法提取匙羹藤总皂苷的最佳工艺为:酶解温度50℃,酶解时间3.0h,酶用量为8mg纤维素/g样品,p H5.5,固液比1∶10条件下,匙羹藤总皂苷提取率最高。结论此最佳工艺可用于匙羹藤茎总皂苷的提取。Objective Through orthogonal experiment of cellulase extraction optimum extraction process of the total saponins of Gymnema sylvestre.Methods Total saponins of Gymnema sylvestre has the chromogenic reaction in the mixture system of 5% vanillin- glacial acetic acid and perchloric acid.The total saponins has the maximum absorption at 500 nm.The total saponin content measured by colorimetric method for Investigation index of total saponins yield.To determine the optimum extraction process through single factor test and orthogonal test by using enzyme- alcohol extraction,butanol extracting and using the total saponins of Gymnema sylvestre extract rate as evaluation indicator.Results The optimum extraction process of total saponins of Gymnema sylvestre is when the enzymolysis temperature is 50℃,hydrolysis time is 2.5 h,the amount of enzyme is 8mg cellulose / g samples,p H5.5,ratio of solid to liquid is under 1： 15,total saponins of Gymnema sylvestre has the highest extraction rate.Conclusion The optimum process is applicable for extraction of the total saponins of Gymnema sylvestre.
