缺血性脑卒中中医证型与TNF-α基因G238A和G308A多态性的相关性研究

摘　　要：目的探讨广西汉族人群不同中医证型缺血性脑卒中易感性与肿瘤坏死因子-α(TNF-α)G238A和G308A位点基因多态性的关系。方法采用Sequenom Mass ARRAY中通量分型方法对255例风痰瘀阻证以及182例气虚血瘀证缺血性脑卒中患者的G238A和G308A位点进行基因分型检测,并对其基因型及基因频率进行比较。结果风痰瘀阻证以及气虚血瘀证患者与对照者间的TNF-α(G238A,G308A)基因型及基因频率分布的差异无统计学意义(P>0.05)。结论TNF-α基因G238A和G308A多态性与缺血性中风之风痰瘀阻证及气虚血瘀证的易感性可能无关。Objective To estimate the distribution TNF- α of gene polymorphism genotype and allele frequencies on ischemic stroke（ IS） Guangxi Han patients with different traditional Chinese medicine（ TCM） syndrome.Methods 437 ischemic stroke cases（ 255 Feng Tan Yu Zu and 182 Qi Xu Xue Yu syndrome samples） and 612 healthy controls were recruited at the same period of time.A Sequenom Mass ARRAY genotyping method was carried out to analyze the 2 single nucleotide polymorphisms： TNF- αgene variant rs1800629（- 308 G / A） and rs361525（- 238 G / A） were detected in our study.Statistical analyses were carried out with SPSS16.0 for windows.Results No statistically significant associations between TNF- α（- 238 G / A and- 308 G / A） gene polymorphisms and the risk of Guangxi Han Feng Tan Yu Zu syndrome ischemic stroke patients were found.In addition,the rs1800629 and rs361525 polymorphisms were not statistically significant associated with Guangxi Han Qi Xu Xue Yu syndrome ischemic stroke patients.Conclusion The TNF- α- 238 G / A and- 308 G / A gene polymorphisms may be not a susceptible predictor of Guangxi Han Feng Tan Yu Zu and Qi Xu Xue Yu syndrome ischemic stroke patients.
