鸦胆子油乳对肺鳞癌细胞增殖的抑制作用

摘　　要：目的探讨鸦胆子油乳诱导人肺鳞癌细胞株SK-MES-1的凋亡作用和其抗肿瘤机制。方法培养人肺鳞癌细胞株SK-MES-1,用不同浓度的鸦胆子油乳处理SK-MES-1细胞,采用MTT法检测鸦胆子油乳对SK-MES-1细胞生长、增殖的抑制作用;流式细胞术检测细胞凋亡率及细胞周期。结果鸦胆子油乳抑制SK-MES-1细胞增殖和诱导SK-MES-1细胞的凋亡作用均呈剂量和时间依赖性。与对照组相比,鸦胆子油乳处理后的SK-MES-1细胞G0-G1期百分比明显增高,S期细胞比例下降。结论鸦胆子油乳在体外能够抑制人肺鳞癌细胞株SK-MES-1的增殖,诱导其细胞凋亡,诱导作用具有显著的量效及时效关系。Objective To investigate the proapoptotic effect and mechanism of javanica oil emulsion on human lung squamous carcinoma SK-MES-1 cells.Methods Cultured human lungs squamous carcinoma SK-MES-1 cells were treated with different concentrations of javanica oil emulsion.Inhibitory effect of javanica oil emulsion on the proliferation of SK-MES-1 cells was detected by MTT.Flow cytometry was performed to examine the cell apoptotic rates and cell cycle profiles.Results The effect of javanica oil emulsion on proliferation and apoptosis on SK-MES-1 cells were in dose- dependent and time-dependent manners.Compared to the control,javanica oil emulsion treatment led to a significant increase of percentage in G0/G1 phase and decrease of percentage in Sphase.Conclusion Javanica oil emulsion can effectively inhibit growth and induce apoptosis of human lung cancer SK-MES-1 cell in vitro,which shows obvious dose- dependent and time-dependent manners.
