摘  要:目的研究亚砷酸(As2O3)对人鼻咽癌CNE-2裸鼠移植瘤与survivin、PTEN的作用,探讨该药物的抗肿瘤机制。方法建立人鼻咽癌裸鼠移植瘤模型,随机分为Na Cl组、As2O3组和DDP组,观察裸鼠移植瘤生长情况;采用免疫组织化学方法检测survivin、PTEN的表达。结果 As2O3组和DDP组瘤体积和瘤重均明显低于Na Cl组,survivin蛋白的表达均低于Na Cl组,PTEN蛋白的表达均高于Na Cl组,差异具有统计学意义(P<0.05)。结论亚砷酸可抑制人鼻咽癌CNE-2裸鼠移植瘤的生长,此作用可能与下调survivin、上调PTEN的表达相关。Objective To study the effects of arsenic trioxide(As2O3) on the growth of human nasopharyngeal carcinoma cell strain CNE-2 xenograft in nude mice and the expression of survivin and PTEN, and explore the antitumor mechanisms of the drug. Methods The models of human poorly differentiated nasopharyngeal carcinoma in nude mice were established and randomly divided into NaCl group,As2O3 group and DDP group. The growth of xenograft in nude mice was observed, and the expressions of survivin and PTEN were detected by immunohistochemistry. Results The tumor volume and weight of As2O3 group and DDP group were significantly lower than those of NaCl group, the expression of survivin of As2O3 group and DDP group was significantly lower than that of NaCl group, the expression of PTEN of As2O3 group and DDP group was significantly higher than that of NaCl group, all the difference was statistical(P〈0.05).Conclusions As2O3 can inhibit the growth of human nasopharyngeal carcinoma cell strain CNE-2 xenograft in nude mice, the mechanism of which might be related with down-regulation of survivin and up-regulation of PTEN.
