缺血性脑卒中大鼠模型海马组织中的NO和H_2S的检测与意义

摘  要:目的研究缺血性脑卒中大鼠模型海马组织中的一氧化氮(NO)和硫化氢(H2S)的动态变化,从而有助于了解NO/H2S与缺血性脑卒中的关系。方法120只SD大鼠被随机分为正常对照组和缺血性脑卒中模型组,每组各60只。比较两组不同时间点平均临床评分、H2S和NO水平。采用苏木素-伊红(HE)染色观察各组脑组织病理变化,分光光度法检测脑组织匀浆的H2S和NO水平。结果模型组术后1天、3天、5天、10天、15天、20天的临床评分均显著高于正常对照组(P<0.05)。大鼠海马组织中H2S水平术后第1天开始升高,第3天达到高峰,随后模型组H2S水平呈逐渐下降趋势,在第20天达到最低,第15天开始与正常组比较已经无统计学差异(P>0.05)。大鼠海马组织中NO水平术后第1天开始升高,第3天达到高峰,随后模型组NO水平呈逐渐下降趋势,在第20天达到最低,第20天与正常组比较已经无统计学差异(P>0.05)。相关性分析显示模型组中大鼠海马组织中H2S水平的变化与临床评分、NO水平呈正相关,相关系数分别为r=0.78、0.87,临床评分和NO水平呈正相关(r=0.82)。结论 H2S和NO在缺血性脑卒中模型海马组织中水平较健康对照组明显增高,与临床评分呈正相关。Objective To study the dynamic change of nitric oxide(NO) and hydrogen sulfide(H2S) in hippocampus tissue of rat models of ischemic stroke, thus helping us to understand the relationship between NO/H2 S and ischemic stroke. Methods 120 SD rats were randomly divided into control group and model group of ischemic stroke, with 60 rats in each group. The average clinical score and the level of H2 S and NO at different time between two groups were compared. The pathological change of brain tissue in different groups was observed by HE staining, the level of H2 S and NO in brain tissue homogenate were detected by spectrophotometric method. Results The clinical scores 1 d, 3 d, 5 d, 10 d, 15 d and 20 d after operation of model group were higher than those of control group(P〈0.05). The H2 S level in hippocampus tissue of model group was increased at 1 d and decreased at 3 d(the level at 3 d was the highest value), and achieved the lowest value at 20 d, which had no statistical difference compared with control group at 15 d(P〉0.05). The NO level in hippocampus tissue of model group was increased at 1 d and decreased at 3 d(the level at 3 d was the highest value), and achieved the lowest value at 20 d, which had no statistical difference compared with control group at 20 d(P〉0.05). The correlation analysis showed that the change of H2 S level in hippocampus tissue of model group was positively related with clinical score(r = 0.78) and NO level(r = 0.87), the clinical score was positively related with NO level(r = 0.82). Conclusions The levels of NO and H2 S in hippocampus tissue of rat models of ischemic stroke are higher than those of healthy rats, which has positive correlation with clinical score.
