SGX523对舌鳞癌Tca8113细胞增殖与c-Met表达的影响

摘　　要：目的：体外观察 SGX523对舌鳞癌细胞株 Tca8113中 c-Met 的表达情况及细胞增殖的影响。方法实验分为2组：对照组（Tca8113＋DMSO）；SGX 组（Tca8113＋不同浓度范围30～600 nmol· L－1的 SGX523）。采用Western blotting 检测不同浓度的 SGX523作用下的 Tca8113细胞中 c-Met、丝裂原活化蛋白激酶（MAPK）和 AKT的表达情况，MTT 法检测不同浓度的 SGX523对 Tca8113细胞作用24、48、72 h 后细胞增殖的抑制率。结果与对照组相比，SGX 组在相同时段的 Tca8113细胞的增殖抑制率明显增加（P ＜0．01），表明 SGX523对 Tca8113细胞的增殖抑制作用具有浓度依赖性。在相同浓度（100 nmol·L－1）SGX523作用下，24、48、72 h 的半数抑制率 IC50分别为（60．31±7．11）、（26．71±4．54）、（7．87±4．92）nmol·L－1，组间比较差异有统计学意义（P ＜0．01），表明SGX523对 Tca8113细胞的增殖抑制作用有时间依赖性。对照组细胞抑制率在24、48、72 h 差异均无统计学意义（均 P ＞0．05）。Western blotting 法检测结果显示不同浓度 SGX523的 SGX 组 c-Met、MAPK 和 AKT 蛋白表达都明显下调，与对照组比较差异有统计学意义（均 P ＜0．05），且不同浓度 SGX523的 SGX 组间比较差异有统计学意义（P ＜0．01）。结论SGX523具有以浓度和时间依赖性方式抑制 Tca8113细胞增殖和下调 c-Met 蛋白表达作用，是一种有前景的舌鳞癌治疗的药物。Objective To observe the effects of SGX523 on the proliferation of tongue squa-mous cell carcinoma Tca8113 cells and the expression of c-Met in vitro.Methods Tca8113 cells were treated with DMSO （control group）or in combination with different concentrations of SGX523 （SGX group,30-600 nmol·L-1 ）.The expression of c-Met,MAPK and AKT was detec-ted by Western blotting.The proliferation of Tca8113 cells was measured after treatment for 24, 48 and 72 hours by MTT assay.Results Compared with control group,SGX523 treatment signif-icantly inhibited the proliferation of Tca8113 cells in a concentration-dependent manner （P 0.05 ）.These results showed that SGX523 inhibited the proliferation of Tca8113 cells in a time-dependent manner.In addition,compared with control group,SGX523 treatment obviously decreased the expression of c-Met,MAPK and AKT protein in a concentra-tion-dependent manner （P 〈0.05 or P 〈0.01）.Conclusion SGX523 treatment can inhibit the proliferation of Tca8113 cells and decrease the expression of c-Met in a time-and concentration-dependent manner.Therefore,SGX523 is a promising drug for tongue squamous cell carcinoma.
