摘 要:目的:探讨RNA干扰技术对脑缺血大鼠的治疗作用。方法:将大鼠随机分为空白对照组、阴性对照组和实验组,携带肾素(renin,REN)基因特异短发夹RNA(short hairpin RNA,shRNA)编码序列的质粒载体与脂质体按1∶4复合,经尾静脉注射到大鼠体内,再对每组大鼠制作缺血再灌注模型;缺血2 h再灌注24 h,记录神经功能评分,检测REN mRNA的表达水平和大鼠血清中REN浓度。结果:神经功能评分提示实验组大鼠损伤较轻(P<0.05),RT-PCR试验提示实验组基因序列表达减少,实验组大鼠血清REN含量减少。结论:成功通过质粒载体介导RNA干扰抑制REN基因的表达,认为RNA干扰技术对脑缺血再灌注大鼠有治疗作用。Objective: To explore the therapeutic effect of RNA interference technology for cerebral ischemia in rats. Methods: Rats were randomly divided into blank control group,negative control group and the experimental group,and then plasmid vector and liposome with renin( REN) gene specific short hairpin RNA( shRNA) coding sequence compounded by the ratio of 1∶ 4 were injected into rats through tail vein. The model of reperfused rats with cerebral ischemia was created,and neurological severity scores were recorded after 2 hours' ischemia and 24 hours' reperfusion,with REN-mRNA expression level and serum REN concentrations detected. Results: Nerve function score suggested the rats' damage in the experimental group was lighter( P〈0. 05). RT-PCR found the expression of gene sequence and the content of serum REN in rats decreased in the experimental group. Conclusions: REN gene expression of successful plasmid vector-mediated RNA interference suppression indicates that RNA interference technology has a treatment effect on reperfused rats with cerebral ischemia.
