摘　　要：目的探讨肿瘤干细胞标记物Nestin对肿瘤细胞抗凋亡的影响。方法采用骨肉瘤细胞MG63细胞株,经过0.9μmol/L顺铂处理12 h、洗涤后经免疫组化染色,显微镜观察幸存细胞的特性;并通过彗星实验验证0.9μmol/L顺铂对Nestin表达阳性细胞和Nestin表达阴性细胞的损伤程度差异。结果显微镜观察显示,顺铂处理后会有少量细胞幸存,且这些未凋亡的细胞绝大多数为Nestin表达阳性细胞;采用彗星实验的研究表明,Nestin表达阳性细胞经顺铂处理后的细胞核染色体损伤较Nestin表达阴性细胞更小。结论 Nestin可能在骨肉瘤MG63细胞抵抗化疗药物顺铂诱导的DNA损伤中起细胞核保护作用。Objective To investigate the effect nestin, a marker of tumor stem cells, on the anti - apoptosis mechanism of in tumor cells. Methods The osteosarcoma cell line MC,63 was utilized for the following investigations. After 0. 9 μ mol/L cisplatin treatment for 12 h and immunohistochemistry （IHC） staining, the characteristics of surviving cells were observed under the microscope. Damage of 0. 9 p, mol/L eisplatin treatment on nestin - positive cells and nestin - negative cells were compared using comet assay. Results Microscopic observation showed that a small amount of cells survived after cisplatin treatment, the majority of which were nestin - positive. Comet assay demonstrated that damage of cisplatin treatment on nuclear chromosomes in nestin - positive cells was weaker than in nestin - negative cells. Conclusion In the osteosarcoma cell line MG63, nestin may exert a protective role on the nucleus and provide chemotherapy resistance in cisplatin- induced DNA damage.
