摘  要:目的探讨骨形态发生蛋白4(bone morphogenetic protein 4,BMP4)在β淀粉蛋白(amyloidβ,Aβ)诱导的神经毒性病变中的变化。方法 SD大鼠海马神经元原代培养,经过不同浓度(10、15、20μmol/L)Aβ25-35毒性片段处理后,通过定量PCR技术检测BMP4mRNA表达水平。SD大鼠双侧基底前脑注射Aβ1-40建立大鼠痴呆模型,通过免疫组化及免疫印迹检测基底前脑及海马区BMP4蛋白表达水平。结果 Aβ25-35处理组海马神经元内BMP4mRNA表达较空白对照组明显减少(P<0.05)。免疫组化和免疫印迹实验结果显示,与对照组比较,Aβ1-40注射组大鼠基底前脑区BMP4蛋白表达减少,海马区其表达升高(P<0.05)。结论 Aβ直接下调神经元内BMP4转录及表达,不同脑区内BMP4蛋白可能存在负反馈调节作用。Objective To investigate the change of bone morphogenetic protein 4(BMP4)expression in amyloid β(Aβ)-induced neurotoxicity. Methods Hippocampal neurons were treated with different concentrations of Aβ25-35(10,15,20μmol/L)for 24 h.The mRNA level of BMP4 in rat hippocampal neurons after Aβ25-35 treatment was assayed through realtime PCR.In vivo,Aβ1-40 was bilaterally injected into basal forebrain to simulate neuropathological processes of AD,and the levels of BMP4 protein were tested in the regions of basalforebrain and hippocampus.Results Results of realtime PCR showed that the levels of BMP4 mRNA in Aβgroups were significantly decreased compared with the control group.Immunohistochemisty and western blot results showed that BMP4 protein was significantly decreased in the Aβgroup relative to the control group in basalforbrain,while there was an opposite change in hippocampus.Conclusions Aβdirectly downregulates BMP4 expression,and there may be anegative feedback regulation of BMP4 in different brain regions.
