褐藻糖胶对多发性骨髓瘤细胞株RPMI8226中Wnt/β-catenin通路的影响研究

摘  要:目的探讨褐藻糖胶诱导多发性骨髓瘤细胞株RPMI8226凋亡过程中Wnt/β-catenin通路的变化。方法应用褐藻糖胶诱导多发性骨髓瘤细胞株RPM18226的凋亡,观察多发性骨髓瘤细胞株经10,25,50,100μg/m L浓度褐藻糖胶分别作用24,48,72 h后细胞增殖抑制率。将细胞分为3组:空白组不予药物干预,褐藻糖胶组给予25μg/m L褐藻糖胶培养,Wnt通道抑制剂(DKK-1)组给予DKK-1培养。用Annexin V-FITC/PI双染法检测3组RPMI822细胞凋亡率,通过Western blot方法检测3组β-catenin与c-myc蛋白表达情况。结果随褐藻糖胶浓度的增高和作用时间的延长,细胞增殖抑制率明显增高(P均<0.05)。褐藻糖胶组细胞凋亡率明显高于空白组;褐藻糖胶组和DKK-1组β-catenin和c-myc蛋白表达水平均明显低于空白组(P均<0.05),褐藻糖胶组与DKK-1组比较差异无统计学意义(P均>0.05)。结论褐藻糖胶能够诱导多发性骨髓瘤细胞株RPMI8226发生凋亡,与DKK-1作用相当,并且褐藻糖胶能够有效抑制多发性骨髓瘤细胞株中Wnt/β-catenin通路的活化状态,为有效治疗多发性骨髓瘤患者提供了理论依据。Objective It is to investigate the changes of Wnt / β-catenin pathway in apoptosis of fucoidan induced multiple myeloma cell line RPMI8226. Methods The apoptosis of multiple myeloma cell line RPMI8226 was induced by fucoidan,then the inhibiting rate of proliferation of the cells were observed after induction for 24,48 and 72 h with 10,25,50,100 μg /m L of fucoidan. The cells were divided into 3 groups: blank group was given no intervention of medicine,fucoidan group was given 25 μg / m L for culture,Wnt channel inhibitor( DKK-1) group was given DKK-1 for culture. Apoptosis rate of RPMI822 cells in the three groups were detected by Annexin V-FITC / PI double staining method,and the expression of β-catenin and c-myc protein was detected by Western blot method. Results With the increase of concentration and effect time of Fucoidan,the inhibition rate of proliferation of multiple myeloma cells increased signficantly( P〈0. 05). The apoptosis rate of fucoidan group was obviously higher than that in the blank group; the levels of expression of β-catenin and c-myc protein in fucoidan group and DKK-1 group were significantly lower than that in blank group( P〈0. 05),but there was no significant difference in the levels between fucoidan group and DKK-1 group( P〈0. 05). Conclusion Fucoidan can induce the apoptosis of multiple myeloma cell line RPMI8226,the effect is similar to DKK-1,and it can effectively inhibit the activation status of Wnt / β-catenin pathway,this may provide some theoretical evidences for the treatment of multiple myeloma.
