摘  要:目的分析长链非编码RNA MEG3及其内含子miRNA-770-5p在上皮性卵巢癌中的表达及其意义,为探讨上皮性卵巢癌的发生机制提供新的思路和方向。方法收集2008-01-06-2013-08-29南京医科大学第一附属医院妇科收治并经病理确诊的卵巢良性肿瘤43例(对照组),上皮性卵巢癌62例(实验组)。应用实时定量聚合酶链反应(polymerase chain reaction,PCR)检测实验组和对照组中MEG3、miR-770-5p及其靶基因ARPC2的相对表达量,并利用Pearsonχ2相关分析法分别分析其相关性,采用χ2检验分析上皮性卵巢癌患者中MEG3的表达量与临床病理资料的关系。结果上皮性卵巢癌组织MEG3表达为0.091±0.012,低于卵巢良性肿瘤组的0.246±0.027,t=5.541,P<0.001;上皮性卵巢癌组织miR-770-5p表达为0.036±0.006,低于卵巢良性肿瘤组的0.251±0.042,t=6.077,P<0.001;上皮性卵巢癌组织靶基因ARPC2表达为0.299±0.034,高于对照组的0.060±0.012,t=5.639,P<0.001。MEG3与miR-770-5p之间的表达呈正相关,r=0.469,P=0.003;而miR-770-5p与靶基因ARPC2的表达呈负相关,r=-0.663,P<0.001。MEG3的表达与上皮性卵巢癌患者淋巴结转移有关,χ2=3.387,P=0.034;而与患者的年龄、临床病理分期、病理分级和组织学类型无关,P>0.05。结论 lncRNA MEG3与其内含子microRNA miR-770-5p在上皮性卵巢癌中表达下调而靶基因ARPC2上调,相互之间呈一定相关性,提示它们可能在上皮性卵巢癌的的发生与进展中起重要作用。OBJECTIVE To explore the expressions of lncRNA MEG3 and its intronic microRNA miR-770-5p in patients with epithelial ovarian cancer(EOC)and provide new insights and directions for the study of the mechanism of EOC.METHODS Collecting tissue samples confirmed by pathology in Gynecology Department,First Affiliated Hospital of Nanjing Medical University from 2008-01-06 to 2013-08-29,43 cases of benign ovarian tumors as control group,62 cases of epithelial ovarian cancer as experimental group and real-time quantitative PCR(polymerase chain reaction)was used to detect the expressions of MEG3,miR-770-5p and its target gene ARPC2 both in the experimental and the control groups and the relevance was revealed using Pearsonχ2 correlation analysis.χ2 test was used to analyze the relationship between levels of MEG3 expression and clinical data in patients with EOC.RESULTS Compared with the control group,the expressions of MEG3 and miR-770-5p in EOC group were significantly lower(0.091±0.012,0.246±0.027,t=5.541,P〈0.001;0.036±0.006,0.251±0.042,t=6.077,P〈0.001).while the expression of the target gene ARPC2 was opposite(0.299±0.034,0.060±0.012,t=5.639,P〈0.001).MEG3 expressions were positively correlated with miR-770-5p(P=0.003,r=0.469),while miR-770-5p and target gene ARPC2 expressions were negative correlation(P〈0.001,r=-0.663).MEG3 expressions in patients with EOC were related with lymph node metastasis(χ2=3.387,P=0.034).Patient's age,clinical staging,pathological grade,histological type were not statistically significant.CONCLUSION We conclude that the expressions of lncRNA MEG3 and its intronic microRNA miR-770-5p were down-regulated while the target gene ARPC2 was up-regulated,showing a certain correlation among them,and such association indicated that LncRNA MEG3 may be involved in the tumorgenesis and progression of EOC by the way of regulating intronic microRNA miR-770-5p.
