摘 要:目的:观察云连总生物碱对小鼠焦虑行为的影响,并探讨其作用机制。方法:60只雄性SPF级昆明种小鼠,随机分成5组,每组12只,分别为模型组,地西泮组,云连总生物碱高剂量组、中剂量组、低剂量组。在高架十字迷宫和明暗箱装置中,观察云连总生物碱对小鼠焦虑行为的影响,然后断头取脑测定脑内γ-氨基丁酸(gamma-aminobutyric acid,GABA)、谷氨酸(glutamic acid,Glu)和5-羟色胺(5-hydroxytrypatmine,5-HT)的含量,并对行为学实验数据和脑内递质含量作统计分析。结果:在高架十字迷宫和明暗箱装置中,云连总生物碱中剂量组对焦虑小鼠有显著抗焦虑作用(P<0.05);小鼠脑内GABA含量统计结果显示,与模型组比较,地西泮组和云连总生物碱中剂量组小鼠脑内GABA含量显著增高(P<0.05),高剂量组和低剂量组有增高趋势,而Glu和5-HT在各组中差异均无统计学意义。结论:云连总生物碱对小鼠有明显抗焦虑作用,以中剂量作用明显。其作用机制与明显升高小鼠脑内抑制性递质GABA的含量有关。Objective:To observe the effect of the total alkaloids of the Yunlian on the anxiety in mice to investigate its mechanism.Meth-ods:60 male SPF Kunming mice were randomly divided into 5 groups,1 2 rats in each group,respectively as model group,Diazepam group,Yunlian total alkaloids in high dose group,middle dose group and low dose group.In the elevated plus maze and clear black box device,observed the Yunlian total alkaloids on anxiety behavior in mice and detected the brain gamma-aminobutyric acid(GABA),glut-arnic acid(Glu)and 5-hydroxytrypatmine(5-HT)content,and the behavior of the experimental data and the brain in neurotransmitters were statistically analyzed.Results:In the elevated plus maze and clear black box device,Yunlian total alkaloid middle dose group had significant anti anxiety effect(P 〈0.05).Compared with the model group,mice brain GABA content of Diazepam group and Yunlian to-tal alkaloid had increased significantly(P 〈0.05)from Mouse brain GABA content statistical results.High dose group and low dose group had increasing trend,and Glu and 5-HT in each group had no statistical significance.Conclusion:The total alkaloid of the Yunlian has obvious anti anxiety effect in mice,and the middle dose of the total alkaloids is obvious.The mechanism of its action is related to the contents of inhibitory neurotransmitter GABA in brain of mice.
