摘 要:目的研究d-δ-三烯生育酚对人晶状体上皮SRA细胞增殖和凋亡的影响,探讨后发性白内障治疗的新方法。方法将人晶状体上皮SRA细胞分为对照组和实验组,对照组不加药物干预,实验组设3个药物浓度,分别为30μmol·L-1、40μmol·L-1、50μmol·L-1的d-δ-三烯生育酚,药物干预人晶状体上皮SRA细胞24 h和48 h后,采用MTT法、Hoechst33258荧光染色和流式细胞仪分析人晶状体上皮SRA细胞的增殖和凋亡情况。结果 d-δ-三烯生育酚呈浓度-时间依赖性抑制人晶状体上皮SRA细胞的增殖,诱导其凋亡。30μmol·L-1、40μmol·L-1、50μmol·L-1的d-δ-三烯生育酚干预48 h对SRA细胞的增殖抑制率分别为(32.23±5.25)%、(54.17±1.63)%和(86.80±0.85)%,与对照组比较差异均有统计学意义(均为P<0.05);干预24 h增殖抑制率分别为(12.70±1.20)%、(19.53±0.95)%和(51.83±6.11)%,与对照组相比差异均有统计学意义(均为P<0.05)。干预24 h后,各实验组细胞晚期凋亡率Q2分别为(5.87±0.15)%、(11.37±0.85)%、(22.77±2.41)%,与对照组的(1.10±0.17)%相比,差异均有统计学意义(均为P<0.05);各实验组细胞早期凋亡率Q4分别为(3.57±0.32)%、(4.20±0.40)%、(8.00±0.50)%,与对照组的(2.53±0.15)%相比,差异均有统计学意义(均为P<0.05)。结论 d-δ-三烯生育酚抑制人晶状体上皮SRA细胞增殖,诱导其凋亡,具有抗晶状体上皮细胞增殖的生物学作用,为治疗后发性白内障提供一种新思路。Objective To study the effects of d-δ-tocotrienols on proliferation and apoptosis of human lens epithelial SRA cell line, and explore a new method of after- cataract treatment. Methods Human lens epithelial SRA cell line was randomly divid- ed into the control group and experimental group, the control group without drug inter- vention, and the experimental group with 3 drug concentrations, respectively 30 μmol L-1 ,40 μmol L-1 ,and 50 μmol L-1 d-δ-tocotrienol. After drug intervention of human lens epithelial SRA cell line for 24 hours and 48 hours, the proliferation and apoptosis of human lens epithelial cells were tested by MTT assay, Hoechst33258 fluorescence stai- ning and flow cytometry. Results d-δ-tocotrienols inhibited the proliferation and ap- optosis of human lens epithelial SRA cell line with a time- and dose-dependent manner. The cell proliferation inhibitive rates of the 30 μmol L-1, 40 μmol L-l, and 50μmol L-1 groups were (32.23±5.25)% , (54.17 ±1. 63)% and (86.80 ±0.85)% ,re- spectively, compared with the control group, the differences were statistically significant (all P 〈 0.05 ). After intervention for 24 hours, the rates of late apoptosis ( Q2 ) were (5.87 ± 0.15 ) % , ( 11.37± 0.85 ) % , ( 22.77± 2.41 ) % , respectively, compared control group ( 1. 10 ±0.17 )% ,the differences were statistically significant ( all P 〈 0. 05 ). The rates of early apoptosis (Q4) were ( 3.57 ± 0.32) % , (4.20 ± 0.40) % , ( 8.00 ± 0.50) % , respectively,compared with control group (2.53 ±0.15 )% ,the differences were statis- tically significant ( all P 〈 0.05 ). Conclusion D-δ-tocotrienols inhibits the proliferation of human lens epithelial SRA cells, induce its apoptosis, and it possesses anti-prolif- eration biological effect on lens epithelial cell, which provides a new idea for the treat- ment of after-cataract.
