MMP-2和MMP-9在食管鳞状细胞癌患者癌组织中的表达及其意义

摘　　要：目的:探讨食管鳞状细胞癌患者癌组织中基质金属蛋白酶2(MMP-2)和基质金属蛋白酶9(MMP-9)表达,分析二者的关联性。方法:选取食管鳞状细胞癌手术离体标本100例,取癌组织和对应的癌旁正常组织,采用逆转录-聚合酶链式反应(RT-PCR)法检测MMP-2和MMP-9 mRNA表达,采用Western blotting法检测MMP-2和MMP-9蛋白表达;采用Pearson相关分析法分析癌组织中MMP-2和MMP-9mRNA表达的相关性。结果:食管鳞状细胞癌组织和癌旁正常组织中MMP-2 mRNA阳性表达率分别为89.0%(89/100)和76.0%(76/100),两者比较差异有统计学意义(χ2=5.853,P=0.016);食管鳞状细胞癌组织和癌旁正常组织中MMP-9mRNA阳性表达率分别为48.0%(48/100)和16.0%(16/100),两者比较差异有统计学意义(χ2=23.529,P=0.000)。MMP-2mRNA阳性表达率与食管鳞状细胞癌的浸润深度有关联(χ2=4.971,P=0.026),与淋巴结转移无关联(χ2=2.127,P=0.145);MMP-9mRNA阳性表达率与食管鳞状细胞癌的浸润深度有关联(χ2=4.421,P=0.035),与淋巴结转移无关联(χ2=0.617,P=0.432)。MMP-2 mRNA与MMP-9mRNA在食管鳞状细胞癌组织中的阳性表达率呈正相关关系(r=0.274,P=0.006)。结论:MMP-2和MMP-9可能参与了食管鳞状细胞癌的发生发展,但在食管鳞状细胞癌淋巴结转移方面可能不发挥关键作用。Objective To explore the expressions of matrix metalloproteinase-2（MMP-2） and matrix metalloproteinase-9（MMP-9）in the tumor tissue of the patients with esophageal squamous cell carcinoma,and to analyze the correlation between the expressions of MMP-2and MMP-9.Methods 100 spicemens of surgical removed esophageal squamous cell carcinoma were chosen.The tumor tissue and the adjacent normal esophagel mucosa tissue were selected.RT-PCR method was used to detect the expressions of MMP-2and MMP-9mRNA in the esophageal squamous cell carcinoma tissue and the adjacent normal tissue.Western blotting method was used to testify the expressions of MMP-2and MMP-9proteins;the correlation between the expressions of MMP-2and MMP-9was analyzed by Pearson correlation analysis.Results The positive expression rate of MMP-2 mRNA in carcinoma tissue was 89.0%（89/100）,the positive expression rate of MMP-2mRNA in adjacent normal tissue was76.0%（76/100）,and there was statistical difference between two groups（χ^2=5.853,P =0.016）.The positive expression rate of MMP-9mRNA in carcinoma tissue was 48.0%（48/100）,the positive expression rate in adjacent normal tissue was 16.0%（16/100）,and there was statistical difference of the positive expression rate between two groups（χ^2=23.529,P=0.000）.There was a positive correlation between the expression of MMP-2mRNA and the tumor invasion depth（χ^2=4.971,P=0.026）,but there was no correlation between the expression of MMP-2mRNA and the lymph node metastasis（χ^2=2.127,P=0.145）.There was a positive correlation between the expression of MMP-9mRNA and the tumor invasion depth（χ^2=4.421,P=0.035）,but there was no correlation between the expression of MMP-9mRNA and lymph node metastasis（χ^2=0.617,P=0.432）.There was positive correlationship between the positive expression rates of MMP-2 mRNA and MMP-9 mRNA in esophageal cancer tissue（r=0.274,P=0.006）.Conclusion The MMP-2and MMP-9may play a very important role in the occurrence and development of esophageal
